A broad-host-range plasmid containing a fusion of the alkA and lacZ genes of Escherichia coil was introduced into various aerobic and facultative gram-negative bacteria-33 species belonging to 19 genera-to study the induction of expression of the alkA gene by alkylating agents. The bacteria included species of the families Enterobacteriaceae, Pseudomonadaceae, Rhizobiaceae, Vibrionaceae, Neisseriaceae, Rhodospirillaceae, and Azotobacteraceae. Results obtained show that all bacteria tested, except Aeromonas hydrophila, Agrobacterium tumefaciens, Hafnia alvei, Rhizobium meliloti, Salmonella enteritidis, Xanthomonas campestris, and those of the genus Rhodobacter, are able to induce the alkA gene of E. coli in the presence of N-methyl-N'-nitro-Nnitrosoguanidine. All these data indicate that the adaptive response to alkylating agents is present in bacterial species of several families and that the Ada box sequence must be widely conserved.

The Rhodobacter capsulatus recA gene has been isolated and sequenced. Its deduced amino acid sequence showed the closest identity with the Rhodobacter sphaeroides RecA protein (91% identity). However, the promoter regions of both R. capsulatus and R. sphaeroides recA genes are only 64% similar. An Escherichia coli-like LexA binding site was not present in the upstream region of the R. capsulatus recA gene. Nevertheless, the R. capsulatus recA gene is inducible by DNA damage in both hetero- and phototrophically growing conditions. The R. capsulatus recA gene is poorly induced when inserted into the chromosome of R. sphaeroides, indicating that the recA gene of both bacteria possess different control sequences despite their phylogenetically close relationship.

A plasmid library of Salmonella typhimurium was used to complement a temperature-sensitive nrdA4 mutant of Escherichia coli. Complementation was obtained with two different classes of plasmids, one carrying the E. coi nds4B-like genes and the second containing an operon encoding a new bacterial ribonucleotide reductase. Plasmids harboring these new reductase genes also enable obligately anaerobic nnilk:Mudl E. coil mutants to grow in the presence ofoxygen. This operon consists oftwo open reading frames, which have been designated nrdE (2,145 bp) and nniF (969 bp). The deduced amino add sequences of the nniE and sniF products include the catalytically important residues conserved in ribonucleotide reductase enzymes of class I and show 25 and 28% overall identity with the Rl and R2 proteins, respectively, ofthe aerobic ribonucleoside diphosphate reductase ofE. coi. The 3' end of the sequenced 4.9-kb frnaent corresponds to the upstream region of the previously published proU operon of both S. typhimwiwn and E. coil, indicating that the nrdEF genes are at 57 min on the chomosomal maps of these two bacterial speces. Analysis ofthe nrdEF andproU sequences demonstrates that transcription ofthe nriEF genes is in the clockwise direction on the S. typhimwiun and E. coi maps.In both eukaryotic and prokaryotic cells, the biosynthesis of deoxyribonucleotides (dNTPs) from the corresponding ribonucleotides is catalyzed by ribonucleotide reductases (RR), with the exception of dflTP, whose synthesis is derived from dCIP and dUDP. RR plays a crucial role in the balanced supply of DNA precursors. Imbalances of the dNTPs have been associated with different genetic effects, besides modifying the sensitivity of cells to DNA-damaging agents (26). Three classes of RR have been described so far (for a review, see reference 35). Class I enzymes are aerobic and exist in higher organisms and in some prokaryotes. Escherichia coli ribonucleoside diphosphate reductase (RDP reductase) is the best known of this group. The nrdA and nrdB genes encode the a and B polypeptide chains, which form the Rl (a2) and R2 (P2) subunits of the RDP reductase. The E. coli RDP enzyme contains a stable radical located at Tyr-122 as an essential component for catalysis (27)
