Expression of the SIRT2 Gene and Its Relationship with Body Size Traits in Qinchuan Cattle (Bos taurus)

Abstract: Silent information regulator 2 (SIRT2) is a member of the sirtuin family of class III NAD (nicotinamide adenine dinucleotide)-dependent protein deacetylases and may regulate senescence, metabolism and apoptosis. The aims of this study were to investigate whether the SIRT2 gene could be used as a candidate gene in the breeding of Qinchuan cattle. Real-time polymerase chain reaction (RT-PCR) results showed that among all types of tissue that were analyzed, the highest mRNA expression levels of the gene were foun… Show more

“…The general linear model (GLM) using SPSS 20.0 software (SPSS, Inc., Chicago, USA) was used for the association analysis between SNPs and selected traits of carcass quality. The linear model was the same as that used in previous publications of our research group [23,39,40]. The statistical linear model was Y ijkm = u+G i = A j +A k +S m +E ijkm , where Y ijkm are the traits measured on each animal; u is the overall mean for each trait; G i is the fixed effect associated with jth genotype; A j is the fixed effect of the jth age; A k is the fixed effect due to the age of the dam; S m is the random effect with the mth sire; and E ijkm is the standard error.…”

“…The DNA content was estimated spectrophotometrically, and diluted to 50 ng/μl. Meanwhile, body measurement traits (BMTs), including body length (BL), withers height (WH), hip height (HH), rump length (RL), hip width (HW), chest depth (CD), and chest circumference (CC), were measured as described previously [10] for association analyses.…”

“…Qinchuan cattle used in this research are an indigenous breed in China, and are known to have good meat quality, adaptability in farming systems, and desirable physical features [7][8][9]. So, it would be valuable to understand the biological function of genes that are associated with carcass characteristics and body or growth traits [10]. In the process of livestock breeding, body measurement and meat quality traits are used as a tool to assess the economic value of animals.…”

“…According to the sequence of the bovine LPL gene (NM_001075120.1), a primer A (sense: 5'-CTTAAACAGGACCTACGATC-3', anti-sense: 5'-CCTTACTAGCAGTGGGAAT-3') was designed to amplify a 585-bp fragment of the LPL gene in exon 6 using the Primer 5 software (Gui et al, 2015).…”
