Expression of Tumor Necrosis Factor-α, Interleukin-1α, Interleukin-6 and Interleukin-10 in Chronic Otitis Media with Bone Osteolysis

Abstract: The purpose of this study was to evaluate the expression of proinflammatory and immunoregulatory cytokines in chronic otitis media. The expression levels of TNF-α, IL-1α, IL-6 and IL-10 were determined by Western blot analysis of tissue samples obtained during ear surgery. The expression levels of TNF-α, IL-1α and IL-6 in cholesteatoma tissues were substantially higher compared to those determined in the granulation tissue. The highest levels of TNF-α, IL-1 and IL-6 proteins were observed in patients with bone… Show more

“…In contrast, the pro-inflammatory cytokines TNF-α, IL-1β and IL-6 were detected, with their secretion being elevated upon stimulation of the cells with both UHMWPE particles and LPS in a time-dependent manner. It is well known that these cytokines are involved in the development of the condition of periprosthetic osteolysis [53]. Thereby, TNF-α seems to play a leading role as it not only promotes bone resorption, but also impairs bone formation by downregulating the gene expression of the extracellular matrix proteins collagen, osteocalcin, alkaline phosphatase and the transcription factor Runx in osteoblasts [54].…”

“…It has been suggested that mast cells may be involved in the bone resorption observed in CSOM. 138,139 Bone resorption is the process by which osteoclasts break down bone and release minerals, resulting in a transfer of calcium from bone fluid to the blood. It is an important aspect of CSOM contributing to many complications of this disease such as ossicular erosion.…”

“…Second, local immunity in middle ear may act in a different manner contrary to systemic immunity affected by obesity. Third, methodologically, transcription level used in this study does not always correlated with protein level and cytokine expression is regulated both at the transcriptional level and translational mechanisms [20]. However this assumption needs further verification.…”
