Expressions of CD274 (PD-L1) and CD47 rReceptors on the sSurface of bBlast cCells in AML pPatients

Abstract: Acute myeloid leukemia (AML) is a genetically heterogeneous clonal disease defined by the proliferation and accumulation of immature hematopoietic cells in the bone marrow and blood. This study aimed to evaluate the CD47 and CD274 (PD-L1) expression in Iraqi AML patients and its role in the disease and evasion of malignant cells from immune system. The study was conducted on 85 patients diagnosed with leukemia, 25 of them were excluded as they had taken chemotherapy for less than a week and because the proport… Show more

“…Interactions between PTPRC + KC/Macrophage subpopulations or CD1c + KC/Macrophage subpopulations with unexpanded and unexpanded CD8 NK-like T cell subpopulations which were largely inhibitory (LILRB1/2, LGALS9, FASLG, TGFB1-TGFBR3, LGALS9-CD47; Figure 8B). These inhibitory interactions may serve to constrain activation and proliferation of CD8 NK-like T cells in the liver allograft, as CD47 is an immune checkpoint inhibitor which delivers a "don't eat me" signal to macrophages (57) and is required for acceptance of hepatocyte transplants in the mouse (58,59). In contrast, interactions between KC/Macrophage subpopulations and expanded CD8 EFF were primarily activating.…”

“…Similarly, interactions between CD8 EXP NK-like T cells and the CD1C + KC/macrophage subpopulation uniquely identified LGALS9-CD47, an immune checkpoint inhibitor, as a predicted receptor-ligand pair (57)(58)(59). In contrast, interactions between KC/Macrophage subpopulations and expanded CD8 EFF were primarily activating, with CD160-TNFRSF14 (LIGHT), ALOX5-ALOX5AP (lipoxygenase pathway), and LTB-LTBR receptor-ligand interactions occurred specifically between all five KC/macrophage subpopulations and CD8 EXP T cells, but not between KC/macrophage subpopulations and CD8 UNEXP T cells (Figure 8B).…”

“…Acute myeloid leukaemia (AML) is a heterogeneous disease defined by the unregulated growth of proliferative progenitor cells that are incapable of terminal differentiation. It is now obvious that numerous genes are often altered in AML (1). However, it is really challenging to identify transformation driver genes within this large number of leukaemia associated disrupted genes.…”
