EZH2 Mediates miR-146a-5p/HIF-1α to Alleviate Inflammation and Glycolysis after Acute Spinal Cord Injury

Abstract: Acute spinal cord injury (ASCI) is a severe traumatic disease of the central nervous system, the underlying mechanism of which is unclear. This study was intended to study the role of EZH2 and miR-146a-5p/HIF-1α in inflammation and glycolysis after ASCI, providing reference and basis for the clinical treatment and prognosis of ASCI injury. We used lipopolysaccharide (LPS) to induce inflammation of microglia, and we constructed the ASCI animal model. qRT-PCR detected the relative expression levels of EZH2, HIF-… Show more

“…Notably, in some cells, miR-138-5p can directly target HIF-1 α and inhibit its expression [ 36 , 37 ]. Studies have also reported that EZH2 can affect the expression of HIF-1 α by downregulated miR-146a-5p [ 38 ]. These results suggest that dysregulation of EZH2/miR-138-5p axis could downregulate HIF-1 α in direct or indirect pathway.…”

“…[ 19 ] Ni et al found that EZH2 could mediate miR‐146a‐5p/HIF‐1 α to attenuate inflammation and glycolysis after acute spinal cord injury (ASCI). [ 20 ] Additionally, studies have shown that mir‐146a‐5p can regulate AKT, STAT, and AMPK. [ 21–23 ] MiR‐146a‐5p is implicated with the imbalance of Th17/Treg induced by bone marrow mesenchymal stem cell‐derived exosomes via targeting IRAK1.…”

“…At the same time, chronic low-grade inflammation (inflamm-aging) conditions the abnormal activation of transcription factors, which leads to changes in the balance of growth factors, chemokines, cytokines, and ROS, resulting in inflammation and cancer [ 38 ]. Therefore, the rational regulation of HIF is beneficial for reducing inflammatory responses after disease injury [ 39 ]. In addition, inflammatory factors and microbial infections activate immune cells and induce glycolysis to occur, as well as activate HIF-1α [ 40 ].…”
