Fabrication of polystyrene microscale porous substrate and its effects on HL‐7702 cells behaviors

Abstract: A novel polystyrene (PS) substrate with microscale porous structure was facilely fabricated by crystalline-controlled casting method using mixed solvent [N,N-dimethylformamide and ethyl alcohol (v/v)] based on the nonsolvent induced phase separation process. The substrate surfaces exhibited a bi-continuous microscale porous morphology with high porosity, large pore size and pore-pore connection structure. Moreover, behaviors of the normal human liver cell line (HL-7702) seeded on this substrate surface were ca… Show more

“…Human normal liver cells (HL-7702) were obtained from the Institute of Biochemistry and Cell Biology (SIBS, CAS, Shanghai, China) and used in assays at passage 7–25. The cell line has been widely used as a representative model of mammalian liver cells [36] [38] . HL-7702 cells were maintained in RPMI-1640 medium (HyClone) supplemented with 10% FBS (Boster) and 1% penicillin/streptomycin (Solarbio) at 37°C in a 5% CO 2 humidified cell culture incubator.…”

“… 22 As the cells adhere to the micropattern, internal mechanical stresses likely act to differentiate the cell, working to match features to function. 23 , 24 However, while sequestering the cells with microchannels may be beneficial to guide cells, the reduction in cell spreading and change in morphology may also generate genetic abnormalities (such as forming micronuclei), 25 , 26 making cell culture platforms with relevant length scales larger than the cells still more desirable. With this quality in mind, casting and embossing remain two of the more reproducible means to produce PS parts, especially on the industrial scale with injection molding equipment, and remain the method of choice for consumer and industrial parts.…”
