Factors Contributing to Intra-Individual Variation of Serum Constituents: 4. Effects of Posture and Tourniquet Application on Variation of Serum Constituents in Healthy Subjects

Abstract: We studied the effects on 18 serum constituents of posture and prolonged tourniquet application. The subjects were 11 healthy men, ages 20-25 years. The assays were performed on the AutoChemist Multi-Channel Analyzer (AutoChem Instrument AB, Lidingö, Sweden). To compensate for the within-hour variation in these constituents, we drew blood samples at 1100 h and 1130 h on several days. The 1100-h sample was taken after the subjects had been sitting erect for 60 min. The 1130-h sample followed different posture … Show more

“…This explains the drop of these parameters. This is consistent with the observations in a number of studies dealing with the circadian rhythm of proteins and blood volume indices [Statland et al, 1974;Humphrey et al, 1977;Touitou et al, 1986;Haus, 19941. When we examined the profiles of the lymphocyte subtypes, we found that those of CD4, CD3, CD8, and €3 cells were similar to that of total lymphocytes, but the NK cell count, on the other hand, was highest around 16:OO. The consequences of long-term exposure in humans are unknown.…”

“…Specimens of venous blood were collected in the morning after an overnight fasting of 12 hours. In order to avoid preanalytical errors in the serum constituents study, 10 the following protocol was adopted for the collection of blood specimens: 1) supine position for at least an hour; 2) standard cuff sphygmomanometer inflated to a constant pressure of 90 mmHg on the arm from which the specimen was taken; and 3) the blood was taken within a maximum of two minutes into Vacutainer® tubes.…”

“…This could probably explain the lower fetal plasma sodium reported in literature. When analysis is performed on blood sampled after clamping of the cord, the tourniquet effect will within minutes cause a concentration of non-filterable constituents (119). This may explain the higher fetal albumin levels reported in previous studies that analysed blood sampled from the clamped cord.…”
