Factors to consider when interrogating 3D culture models with plate readers or automated microscopes

Abstract: Along with the increased use of more physiologically relevant three-dimensional cell culture models comes the responsibility of researchers to validate new assay methods that measure events in structures that are physically larger and more complex compared to monolayers of cells. It should not be assumed that assays designed using monolayers of cells will work for cells cultured as larger three-dimensional masses. The size and barriers for penetration of molecules through the layers of cells result in a differ… Show more

“…The optimal incubation period should ensure a homogenous diffusion pattern through the microtissue. Weak emitted fluorescence signals in the core of the microtissue might be due to the less viable cells or decreased metabolism of cells in the core [51]. However, the live/dead assay showed viable cells in the internal parts of the microtissues.…”

“…Their size homogeneity was maintained during the subsequent 14-day culture in suspension. When proliferating cells used in 3D culture, the cells located in the external layers show more proliferation, whereas the internal cells in the core area might be quiescent or even necrotic because of limitations in diffusion of O 2 and nutrients [51].…”

“…Drugs usually target certain receptors on the cell surface. The difference in the structure and spatial arrangement of the surface receptors may affect the drug receptor binding efficiency, thereby triggering different responses [73]. Third, cells cultured in 2D are usually at the same cell proliferation stage, while 3D cells are usually at a different cell proliferation stage, similar to cells in the body [72].…”

“…To study the role of EVs in the communication of a cell with its environment, 3D models thus appear a cogent choice. Within the spectrum of available 3D models, one should then consider the complexity, costs, throughput and availability of imaging methods 59 . Yet, depending on the research question, one could still consider 3D models as simplified representations of reality.…”

“…Within the spectrum of available 3D models, one should then consider the complexity, costs, throughput and availability of imaging methods. 59 Yet, depending on the research question, one could still consider 3D models as simplified representations of reality. While it is certainly true that these models can bring crucial insights into the relationship between cellular complexity, space‐geometry, and EV biology, they are commonly not vascularized and/or do not contain stroma as found in tissues within in a living organism.…”
