Fast neurotransmitter release regulated by the endocytic scaffold intersectin

Abstract: Sustained fast neurotransmission requires the rapid replenishment of release-ready synaptic vesicles (SVs) at presynaptic active zones. Although the machineries for exocytic fusion and for subsequent endocytic membrane retrieval have been well characterized, little is known about the mechanisms underlying the rapid recruitment of SVs to release sites. Here we show that the Down syndromeassociated endocytic scaffold protein intersectin 1 is a crucial factor for the recruitment of release-ready SVs. Genetic dele… Show more

“…To explore the physiological functions of ITSN1 in vivo, we analyzed the overall brain architecture of KO mice deficient in ITSN1 (25,26). Nisslstained sections of hippocampi from ITSN1 KO mice showed a gross dispersion of pyramidal neurons in the CA1 and CA3 areas and an altered shape of the dentate gyrus (DG) (Fig.…”

“…Genetic perturbation of endocytic proteins often leads to a delay in vesicle resupply, as shown in mutants of endophilin (54)(55)(56), synaptojanin (57,58), and dap160/intersectin-1 (59). Intersectin-1 KO synapses in mice showed deficient vesicle replenishment even without a detectable change in capacitance decays (60). We tested the vesicle resupply rate by examining the recovery of transmission evoked by an AP at different intervals following each high-frequency stimulation (30 APs at 300 Hz) (Fig.…”
