FcγR Binding and ADCC Activity of Human IgG Allotypes

Abstract: Antibody dependent cellular cytotoxicity (ADCC) is an Fc-dependent effector function of IgG important for anti-viral immunity and anti-tumor therapies. NK-cell mediated ADCC is mainly triggered by IgG-subclasses IgG1 and IgG3 through the IgG-Fcreceptor (FcγR) IIIa. Polymorphisms in the immunoglobulin gamma heavy chain gene likely form a layer of variation in the strength of the ADCC-response, but this has never been studied in detail. We produced all 27 known IgG allotypes and assessed FcγRIIIa binding and ADC… Show more

“…Between donors, however, the IGHG3 * 14 allotype seems to exhibit a higher degree of galactosylation and sialylation in comparison with IGHG3 * 11/12 allotypes. This observation, was also previously described for mAbs by de Taeye et al (11). So far, we only observed 12 of the 27 existing allotypes, with few combinations of them.…”

“…Of note, IgG allotypes were found to correlate with serum IgG levels ( 19 , 43 ) but also with the switching between IgG isotypes suggesting that IgG allotypes may affect the humoral antibody response. Moreover, it was recently demonstrated that allotypic variants within the IgG3 subclass substantially affect FcgRIIIA binding and Antibody-Dependent Cellular Cytotoxicity ( 44 ). To our knowledge, no work has previously shown associations between Gm phenotypes comprising G3m24 or G3m24 alone with a higher risk of malaria infections, while G3m24 is prevalent among sub-Saharan African populations ( 45 , 46 ).…”

“… 8 , 9 Subclass, allotype, and glycosylation, especially fucosylation of IgG impacts FcγR binding. 10 12 FcγR-mediated IgG effector functions are partially regulated by varying FcγR expression on different immune cells. 13 For example, FcγRIIIb or CD16b is exclusively expressed as a monomeric protein mainly on granulocytes, while the homologous FcγRIIIa or CD16a is expressed on NK cells, monocytes, macrophages, and dendritic cells.…”
