FGF-2 Enhances Vascularization for Adipose Tissue Engineering

Abstract: Small intestinal submucosa particles are a favorable scaffold for preadipocytes, allowing ex vivo proliferation on particles small enough to be injected. Delivery of FGF-2 from poly(lactic-co-glycolic acid) microspheres resulted in cell survival and enhanced vascularization.

“…24 Synthetic microsphere encapsulation has also been used to trap bFGF in PLGA, incorporating these microspheres into alginate scaffolds or simply injecting them with small intestinal submucosa and preadipocytes, both of which have been shown to significantly enhance vascularization. 25,26 In terms of biodegradable microspheres, chitosan-gelatin microspheres containing bFGF have been developed and incorporated into a porous chitosan-gelatin scaffold, demonstrating initial fast release kinetics over 2 days before achieving a steady release profile lasting 2 weeks, displaying promise for vascularization. 27 Ultimately, the ability to direct the timing and release of growth factors will control the degree of vascularization within the tissue.…”

“…Bioactive peptide molecules derived from mammalian extracellular matrix (ECM) have been shown to have potent chemoattractive and mitogenic effects upon endogenous progenitor and stem cells (13)(14)(15)(16). Although full characterization of these bioactive peptides has not been completed to date, ex vivo enzymatic, chemical, and physical methods (17)(18)(19), and in vivo physiologic methods (20), have generated a heterogeneous population of ECM peptides (13,21) with both chemotactic and mitogenic properties for a variety of stem and progenitor cells.…”

“…Fibroblast growth factor-2 (FGF-2) and fibroblast growth factor-10 (FGF-10) are also potent inducers of angiogenesis secreted by adipocytes [13,14,26]. Hepatocyte growth factor (HGF) is also secreted by adipocytes and stimulates angiogenesis [3,39].…”
