FH535 Inhibited Migration and Growth of Breast Cancer Cells

Abstract: There is substantial evidence indicating that the WNT signaling pathway is activated in various cancer cell types including breast cancer. Previous studies reported that FH535, a small molecule inhibitor of the WNT signaling pathway, decreased growth of cancer cells but not normal fibroblasts, suggesting this pathway plays a role in tumor progression and metastasis. In this study, we tested FH535 as a potential inhibitor for malignant phenotypes of breast cancer cells including migration, invasion, and growth.… Show more

“…In a previous study [67], H535 at the dose of 15 µmol/L could down-regulate β-catenin expression in HCT-116 and SW48 cells. FH535 at the dose of 0.1 µmol/L also showed to inhibit migration and growth of breast cancer cells [68]. Recent studies have demonstrated that FH535 is a synthetic inhibitor of the canonical WNT-signaling pathway and, not for normal fibroblasts, FH535 also inhibits growth of colon, lung, and hepatocellular carcinoma cell lines [67].…”

“…Glycanated variants concentrate in filopodial extensions, including the finest protrusions conventionally denoted "microspikes," where they seem to colocalize with b1, a3, and a4 integrin subunits, ezrin, and neural precursor cell expressed, developmentally downregulated 9 ( Fig. 4 and Table 1) (17,23,(43)(44)(45)(46)(47). However, NG2/CSPG4 molecules lacking GAG chains have also been noted in microclustered configurations confined to putative lipid rafts.…”
