Fibroblast growth factor-1 attenuates TGF-β1-induced lung fibrosis

Abstract: Idiopathic pulmonary fibrosis (IPF) is characterized by progressive fibroblast and myofibroblast proliferation, and extensive deposition of extracellular matrix (ECM). Fibroblast growth factor-1 (FGF-1) belongs to the FGF family and has been shown to inhibit fibroblast collagen production and differentiation into myofibroblasts, and revert epithelial-mesenchymal transition by inhibiting TGF-1 signalling pathways. However, the precise role of FGF-1 in pulmonary fibrosis has not yet been elucidated. In this stu… Show more

“…Moreover, transient upregulation of FGF1 markedly attenuated TGFβ1-induced lung fibrosis in rats when given either concomitantly or post-injury by enhancing AEC proliferation and hyperplasia 86 . In addition, mice engineered to inducibly overexpress FGF10 in the alveolar epithelium during both the inflammatory phase and the more clinically relevant fibrotic phase of the bleomycin injury model showed a strong attenuation of fibrotic scarring in the lungs 87 .…”

“…Rats were given intratracheal instillations of TGF-β1 adenovirus (Ad-TGF-β1223/225) or control virus (Ad-DL) was prepared and treated to rats as previously described 17. Rats received 5.0×10 8  plaque-forming unit virus in 300 µL sterile saline intratracheally and were culled on day 7, 14, 21 or 35 by terminal anaesthesia.…”

“…These findings taken together thus suggest a critical role for FGF-1 in the development of OA. Interestingly, such a repressive effect of FGF-1 on CCN2 production was strongly implicated by the results of a recent study on lung fibrosis as well (Shimbori et al 2016). According to that study, overexpression of FGF-1 by an adenoviral vector strongly attenuated the TGF-β1-induced lung fibrosis in a rat model in vivo.…”
