FIM-A, a Phosphorus-Containing Sirolimus, Inhibits the Angiogenesis and Proliferation of Osteosarcomas

Abstract: The mTOR pathway is a central control of cell growth, proliferation, metabolism, and survival, and is deregulated in most cancers. Cancer cells are addicted to increased activity of mTOR kinase-mediated signaling pathways, leading to numerous inhibitors of mTOR signaling in preclinic and clinical trials for cancer therapy. Phosphorus-containing sirolimus (FIM-A), which targets mTOR signaling, inhibits cancer cell growth in vitro. Here we report that FIM-A reduces the angiogenesis and proliferation of osteosarc… Show more

“…The materials enriched with P 2 O 5 can quickly bind the calcium ions, accelerating the deposition of calcium phosphates on the composite surfaces, thus reducing the biological activity of free calcium ions [ 25 ]. Furthermore, the correlation between the VEGF expression and phosphorus oxide-containing materials (A2/PLGA and S2/PLGA) could be observed, as CMs from these materials reduced the mRNA levels of VEGF expression, which is consistent with reports showing that phosphorus compounds can affect VEGF expression [ 26 ]. A similar trend could be observed in the case of MMP-2.…”

“…Moreover, Rapamycin can effectively inhibit osteosarcoma stem cells proliferation [22]. Additionally, some moleculars and drugs, such as lupeol, Oleanolic acid, metformin, p53, icariside II, capsaicin, phosphorus-containing sirolimus, heat shock protein 90B1, inhibit OS cell growth and proliferation by targeting AMPK/mTOR and PI3K/AKT/mTOR signaling and down-regulating cyclin D1 and phosphorylation of S6K1 and 4EBP1, which are regarded as downstream target of mTORC1 [23-30]. Besides, overexpression of miR-101 can down-regulate the expression of mTOR, contributing to the inhibition of OS cell proliferation [31].…”

“…Another pathway by which activation of mTOR pathway promotes OS cell metastasis is angiogenesis. P53 and phosphorus-containing sirolimus suppresses OS cell angiogenesis through inhibition of mTOR [26, 30]. Thus, inhibition of mTOR may be a novel effective candidate therapeutic strategy against OS cell metastasis.…”

“…A recent publication from MSKCC also examined the utility of IMRT dose painting in a small group of Ewing sarcoma patients with pulmonary metastasis. There were no local failures, and an overall decreased mean dose to the esophagus, heart, spinal cord, and liver when compared to conventional methods (Yang et al 2013 ). Although the data surrounding IMRT use in Ewing tumors is currently limited, there clearly appears to be an emerging role for this technique in the management of pediatric malignancies.…”

“…Hepatoma-derived growth factor (HDGF) has been shown to be upregulated in Ewing sarcoma and has been reported to be signifi cantly correlated with tumor volume, metastases at diagnosis, low overall survival rate, and low disease-free survival rate (Yang et al 2013 ). It has also been reported to improve prognostic stratifi cation for patients with Ewing family tumors when combined with p53 expression status (Yang et al 2014 ).…”
