First detection ofBursaphelenchus luxuriosaeassociated withPinus pinasterin Portugal and in Europe

Abstract: During a field survey carried out on symptomatic maritime pine trees (Pinus pinaster) in Góis, central Portugal, Bursaphelenchus luxuriosae was isolated for the first time in Portugal, and in Europe. Identification of the nematodes was based on morphological characters and molecular analyses for this species. The general morphology of both females and males is in agreement with the original description for B. luxuriosae, namely the typical morphology of the male spicules and the conspicuous morphology of femal… Show more

“…At the beginning of the 20 th century, PWN was introduced in Japan and became responsible for massive mortality of native pine trees ( Pinus densiflora, P. thunbergii and P. luchuensis ) and then spread to China, Korea and Taiwan3. In Europe, it was first detected in continental Portugal in maritime pine, P. pinaster 4 and more recently in P. nigra 5. It has also been found in Madeira Island6 and Spain78 associated with P. pinaster , representing an increasing threat to European conifer forests.…”

“…Bursaphelenchus luxuriosae described in Japan was identified in Portugal. This was the third member of the xylophilus group in Portugal "It is difficult to ascertain whether B. luxuriosae was introduced, together with its insect vector, or already occurred as a native species (Inácio et al, 2017)." There may be an endemic association of US B. antoniae with another Hylobius in the USA, (Salom, 1997) such as the relatively common pales weevil, H. pales, in eastern North America (www.…”

“…DNA quantity and purity were checked using a NanoDrop 2000 UV-V is Spectrophotometer (Thermo Fisher Scientific, Massachusetts, USA). PCR mixtures, using the primers D2A (5′-ACAAGTACCGTGGGGAAAGTTG-3′) and D3B (5′-TCGGAAGGAACCAGCTACTA-3′) (De Ley et al, 1999) and thermal cycling conditions were performed as described previously by Inácio et al (2016). PCR products were cleaned using the QIAquick PCR Purification Kit (Qiagen, Hilden-Germany), according to the manufacturer's instructions.…”
