Species of Colletotrichum are considered important plant pathogens, saprobes, and endophytes on a wide range of plant hosts. Several species are well-known on citrus, either as agents of pre- or post-harvest infections, such as anthracnose, postbloom fruit drop, tear stain and stem-end rot on fruit, or as wither-tip of twigs. In this study we explored the occurrence, diversity and pathogenicity of Colletotrichum spp. associated with Citrus and allied genera in European orchards, nurseries and gardens. Surveys were carried out during 2015 and 2016 in Greece, Italy, Malta, Portugal and Spain. A total of 174 Colletotrichum strains were isolated from symptomatic leaves, fruits, petals and twigs. A multi-locus phylogeny was established based on seven genomic loci (ITS, GAPDH, ACT, CAL, CHS-1, HIS3 and TUB2), and the morphological characters of the isolates determined. Preliminary pathogenicity tests were performed on orange fruits with representative isolates. Colletotrichum strains were identified as members of three major species complexes. Colletotrichum gloeosporioides s.str. and two novel species (C. helleniense and C. hystricis) were identified in the C. gloeosporioides species complex. Colletotrichum karstii, C. novae-zelandiae and two novel species (C. catinaense and C. limonicola) in the C. boninense species complex, and C. acutatum s.str. was also isolated as member of C. acutatum species complex. Colletotrichum gloeosporioides and C. karstii were the predominant species of Colletotrichum isolated. This study represents the first report of C. acutatum on citrus in Europe, and the first detection of C. novae-zelandiae from outside New Zealand. Pathogenicity tests revealed C. gloeosporioides s.str. to be the most virulent species on fruits. The present study improves our understanding of species associated with several disease symptoms on citrus fruits and plants, and provides useful information for effective disease management.

During the period from 2010 to 2013 preharvest symptoms were detected on different cultivars of sweet orange in six orchards in Catania, Siracusa and Enna provinces, Southern Italy. A total of 56 monosporic fungal isolates were obtained, and among these, 44 were identified as Colletotrichum gloeosporioides and 12 as C. karstii through morphological and molecular analysis. PCR with primers ITS1 and ITS4, primers TubGF1 and TubGR specific for β‐tubulin gene, primers GDF‐GDR, specific for Glyceraldehyde‐3‐phosphate dehydrogenase (GAPDH) gene, were used to confirm the identification of Colletotrichum isolates from citrus. The ITS1‐5.8S‐ITS2 region, a portion of approximately 500 bp of β‐tubulin gene and a fragment of 220 bp of GAPDH gene of the isolates were sequenced and analysed with the BLASTn program. Koch's postulates were fulfilled by pathogenicity tests carried out on fruit of ‘Tarocco Scirè’ and ‘Tarocco Nucellare’ with representative isolates of C. gloeosporioides and C. karstii. Field surveys and pathogenicity tests revealed significant differences in fruit susceptibility between ‘Tarocco Scirè’ and ‘Tarocco Nucellare’ and in virulence between the fungal species. To our knowledge, this is the first report on the emergence of Colletotrichum spp. causing anthracnose in preharvest conditions.

Rumple is a serious peel collapse of Primofiori lemons in the southeast of Spain with an unresolved aetiology. Symptoms typically occur on fruits at ripening under wet conditions as dark sunken lesions producing premature fruit drop and damaged fruits unacceptable for fresh commercialization. A total of 16 Colletotrichum spp. isolates established from rumple-affected lemons collected during the autumn of 2007 from two different orchards were characterized by molecular and phenotypic assays and compared with reference isolates. Species-specific PCR reactions using β-tubulin 2 nucleotide sequences showed Colletotrichum gloeosporioides to predominate (81.5%) with limited occurrence of C. acutatum (18.75%). Among the C. gloeosporioides isolates, five (38.5%) showed benomyl resistance and eight (61.5%) were highly sensitive to the fungicide. The limited occurrence of C. acutatum could be related to factors such as the presence of both species on the same fruit, unfavourable meteorological conditions and low disease incidence. This work reveals an association of C. gloeosporioides and C. acutatum isolates with rumple disease of lemons and expands the range of C. acutatum on citrus.
