FISH-mapping comparison between river buffalo chromosome 7 and sheep chromosome 6: assignment of new loci and comparison with HSA4

Abstract: Synchronized peripheral blood lymphocytes from both river buffalo (BBU) and sheep (OAR) were treated for late incorporation of both BrdU and H-33258 to obtain R-banded preparations to be used for FISH-mapping. Ovine BAC-clones were hybridized for three days on slides pre-exposed to UV light after H-33258 staining. The following loci were mapped: GPR103 (BBU7q13, OAR6q13), TRAM1L1(OAR6q13dist), PPP3CA (BBU7q21, OAR6q15), SNCA (OAR6q17), PPARGC1A(BBU7q23, OAR6q17), UGDH (BBU7q25prox, OAR6q22prox), KDR (BBU7q27, … Show more

“…This study underlines the need of cytogenetic maps for anchoring other maps, such as RH, linkage or sequence contig maps to specific chromosome regions. This way, errors caused by mapping methods based on likelihoods and eventually mismatches in the genome sequence annotation procedure of a species can be avoided [Goldammer et al, 2009b;Perucatti et al, 2009]. Comparison between the present cytogenetic map of OAR1 and the previous RH map [Wu et al, 2009].…”

“…Cytogenetic maps are useful tools for several applications in animal cytogenetics: (a) precise physical position on single chromosome bands of both type I and type II loci, especially using FISH mapping on R-banded chromosome preparations [Hayes et al, 2000;Iannuzzi et al, 2001Iannuzzi et al, , 2003Di Meo et al, 2007; (b) use of molecular markers to confirm chromosomes involved in chromosome abnormalities, as well as detect particular chromosome abnormalities or chromosome regions involved in chromosome abnormalities [reviewed in Ducos et al, 2008 andVillagomez et al, 2009]; (c) studying chromosome abnormalities in germinal cells [Bonnet-Garnier et al, 2008;Villagomez and Pinton, 2008;Vozdova et al, 2008]; (d) anchoring linkage maps, RH maps, and genome sequence contigs to specific chromosome regions [Goldammer et al, 2009b;Perucatti et al, 2009].…”

“…Likewise, the ovine DNA probes that are used should be selected according to their use in comparative mapping approaches, such as cytogenetically mapped loci in other domestic Bovidae such as cattle, goat or river buffalo. Building these bridges between the different maps and chromosome morphology has already been reported for OAR23 [Tetens et al, 2007], OAR10 [Drögemüller et al, 2008], OAR6 [Perucatti et al, 2009] and OARX [Goldammer et al, 2009a, b]. Other map comparisons are being performed for other sheep chromosomes to demonstrate the evolutionary rearrangements in other Bovidae and human.…”

“…Although large-insert DNA libraries from closely related ruminant species such as cattle, goat, and sheep have been used for in situ hybridization experiments in buffalo (Di Meo et al, 2005Perucatti et al, 2009), a buffalo library is necessary for whole-genome physical mapping and isolation of genes and gene clusters as well as for the identification of regulatory elements. Herein we describe the construction of a river buffalo BAC library, a key resource for comparative gene-mapping studies, sequence assembly, and characterization of quantitative trait loci through positional cloning.…”
