Background: Semi-refined carrageenan (food additive E407a) is a widely used thickener, which has been reported to exert toxic and proinflammatory effects. In particular, there is accumulating evidence that it induces eryptosis, i.e. a programmed cell death of eryptocytes, via ROSmediated pathways. However, the role of Ca 2+ -dependent mechanisms in E407a-induced eryptosis is not elucidated.Material and methods: Semi-refined carrageenan at concentrations of 0 mg/ml, 1 mg/ml, 5 mg/ml and 10 mg/ml was incubated with blood of intact female WAG rats (n=9) for 24 h in RPMI and fetal bovine serum. After 24 h, the samples were used to obtain erythrocyte suspensions. The obtained suspensions were stained with a Ca 2+ -sensitive FLUO4 AM probe (30 min, 2.5 µM). The fluorescence of FLUO4 in erythrocytes was detected by a BD FACSCanto II flow cytometer.Results: The intracellular Ca 2+ levels are proportional to the fluorescence of FLUO4. The mean fluorescence intensities (MFI) were compared. Low levels (1 mg/ml) of E407a had no impact on Ca 2+ concentrations in erythrocytes (p>0.05). On the contrary, high concentrations (5 mg/ml and 10 mg/ml) of this food additive promoted an increase in the intracellular Ca 2+ levels. The MFI values were 2.3-and 2.5-fold higher, respectively (p<0.0001). In addition, the exposure to E407a at concentrations of 5 mg/ml and 10 mg/ml (p<0.0001) increased the percentage of cells with high FLUO4 fluorescence.Conclusion: Food additive E407a induces eryptosis in a Ca 2+dependent manner.

Харківський національний медичний університет, Харків, Україна Анотація. Протягом останніх двох десятиліть відмічається глобальне зростання зловживання алкоголем і частоти розладів, пов'язаних з цим. Алкоголь є провідним фактором ризику багатьох видів травм. Симптоми стану відміни алкоголю (СВА) виникають у 31% пацієнтів із травмами. У пацієнтів із політравмою та СВА, ускладненим алкогольним делірієм, відмічають запальний процес, однією з відповідей організму на цей патологічний стан є утворення активних форм кисню (АФК) нейтрофілами. Мета: визначити генерацію АФК в крові пацієнтів із політравмою та СВА з алкогольним делірієм залежно від способу седації. Об'єкт і методи дослідження. Обстежено 80 пацієнтів із політравмою середньої тяжкості та СВА, ускладненим алкогольним делірієм. Медіана віку становила 45 років. Пацієнтам 1-ї групи (n=40) в якості методу седації обрано дексмедетомідин, у 2-й групі (n=40) використовували седацію діазепамом за симптом-тригерним протоколом. Для діагностики делірію використовували метод оцінки сплутаності свідомості у пацієн тів, які перебувають у відділенні інтенсивної терапії (Confusion Assessment Method-Intensive Care Unit -CAM-ICU). Визначення АФК в еритроцитах проводили методом проточної цитометрії із застосуванням програмного забезпечення «BD FACSDiva TM » («Becton Dickinson», США). Результати. У пацієнтів із політравмою та СВА, ускладненим алкогольним делірієм, виявлено значно підвище ну генерацію АФК в еритроцитах, що зумовлює ериптоз. Висновок. Застосування дексмедетомідину може бути рекомендоване в якості седативного засобу в терапії алкогольного делірію у пацієнтів із політравмою та СВА. Ключові слова: політравма, стан відміни алкоголю, алкогольний делірій, дексмедетомідин, активні форми кисню.

Antimicrobial activity and cytotoxicity study of cerium oxide nanoparticles with two different sizes

The control over bacterial diseases requires the development of novel antibacterial agents. The use of antibacterial nanomedicines is one of the strategies to tackle antibiotic resistance. The study was designed to assess the antimicrobial activity of cerium oxide (CeO 2 ) nanoparticles (NP) of two different sizes (CeO 2 NP1 [1-2 nm] and CeO 2 NP2 [10-12 nm]) and their cytotoxicity towards eukaryotic cells. The antimicrobial activity, effects of nanoparticles on DNA cleavage, microbial cell viability, and biofilm formation inhibition were analyzed. The impact of cerium oxide nanoparticles on eryptosis of erythrocytes was estimated using annexin V staining by flow cytometry. The newly synthesized CeO 2 NP1 and CeO 2 NP2 displayed moderate antimicrobial activities. CeO 2 NP1 and CeO 2 NP2 exhibited single-strand DNA cleavage ability. CeO 2 NPs were found to show 100% microbial cell viability inhibition at a concentration of 500 mg/L. In addition, CeO 2 NP1 and CeO 2 NP2 inhibited the biofilm formation of S. aureus and P. aeruginosa. Larger cerium oxide nanoparticles were found to be less toxic against erythrocytes compared with the smaller ones. CeO 2 nanoparticles demonstrate moderate antimicrobial activity and low cytotoxicity towards erythrocytes, which make them promising antibacterial agents.
