Rutin is a flavonoid compound obtained from different vegetables and fruits; specifically, it is found in the seeds of buckwheat and in fruit peels, particularly citrus. It is also an important constituent of red wine. Rutin exhibits various biological properties including antiviral, vasoprotective, anti-inflammatory, and anticarcinogenic activities. However, its antioxidant activity is the most well studied. Despite the potential for in vitro applications, rutin presents low oral bioavailability that affects its biological activities. Nanoparticles composed of polymers, protein, or lipids are of great importance in the pharmaceutical and nutraceutical areas due to their physicochemical properties, which improve the pharmacokinetics of the drug which is loaded within. This study presents the production of bovine serum albumin (BSA) nanoparticles containing rutin by nano spray drying. Nanoparticles were characterized in terms of mean particle size, size distribution, morphology, zeta potential, and drug content; as well as their antioxidant activity. The optimized spray-drying conditions produced spherical particles with a mean size of 316 nm, zeta potential of −32 mV, and encapsulation efficiency around 32%. Moreover, when antioxidant activity toward the ABTS + radical was assayed, nanoencapsulation increased the IC 50 of rutin by 2-fold. The nano spray-drying process proved to be suitable for the production of rutin-loaded BSA nanoparticles with potential antioxidant activity.

Injectable or implantable poly(lactic-co-glycolic acid) (PLGA) devices for the sustained delivery of proteins have been widely studied and utilized to overcome the necessity of repeated administrations for therapeutic proteins due to poor pharmacokinetic profiles of macromolecular therapies. These devices can come in the form of microparticles, implants, or patches depending on the disease state and route of administration. Furthermore, the release rate can be tuned from weeks to months by controlling the polymer composition, geometry of the device, or introducing additives during device fabrication. Slow-release devices have become a very powerful tool for modern medicine. Production of these devices has initially focused on emulsion-based methods, relying on phase separation to encapsulate proteins within polymeric microparticles. Process parameters and the effect of additives have been thoroughly researched to ensure protein stability during device manufacturing and to control the release profile. Continuous fluidic production methods have also been utilized to create protein-laden PLGA devices through spray drying and electrospray production. Thermal processing of PLGA with solid proteins is an emerging production method that allows for continuous, high-throughput manufacturing of PLGA/protein devices. Overall, polymeric materials for protein delivery remain an emerging field of research for the creation of single administration treatments for a wide variety of disease. This review describes, in detail, methods to make PLGA devices, comparing traditional emulsion-based methods to emerging methods to fabricate protein-laden devices. The objective of present study was to prepare positively charged carvedilol-loaded nanoparticles providing a controlled release formulation by spray dryer technique and was to examine the effect of different derivative of Eudragit ® polymers on entrapment efficiency (EE%), dissolution profile and release kinetics. Two nonbiodegradable positively charged polymers, Eudragit ® RS100 and RL100 were used alone or in combination. The prepared formulations were evaluated for their particle size, polydispersity index, zeta potential, in vitro dissolution and in vitro release kinetics study. Particle sizes of placebo nanoparticles and carvedilol loaded nanoparticles were 418.2±18.5/ 402.3±22.0/ 416.3±12.5 and 535.5±13.8/ 529.4±10.2/ 530.4±10.4 nm, respectively with PDI values of approximately 0.4-0.6 for each. Carvedilol loading was resulted in positive electrical charge on nanoparticles. The drug encapsulation efficiency was 68.45±4.67/ 63.58±2.30/ 65.67±4.21. In vitro cumulative release from the nanoparticles was 80-90 % at 48 hour. Morphology and solid state analyzes were performed also in nanoparticles. The results of this research indicate that spray dryer technique is suitable for carvedilol loaded Eudragit ® nanoparticles and no burst effect but a prolonged drug release was observed from formulations.
