The utilization of co-processed excipients (CPEs) represents a novel approach to the preparation of orally disintegrating tablets by direct compression. Flow, consolidation, and compression properties of four lactose-based CPEs—Cellactose® 80, CombiLac®, MicroceLac® 100, and StarLac®—were investigated using different methods, including granulometry, powder rheometry, and tablet compaction under three pressures. Due to the similar composition and the same preparation technique (spray drying), the properties of CPEs and their compacts were generally comparable. The most pronounced differences were observed in flowability, undissolved fraction after 3 min and 24 h, energy of plastic deformation (E2), ejection force, consolidation behavior, and compact friability. Cellactose® 80 exhibited the most pronounced consolidation behavior, the lowest values of ejection force, and high friability of compacts. CombiLac® showed excellent flow properties but insufficient friability, except for compacts prepared at the highest compression pressure (182 MPa). MicroceLac® 100 displayed the poorest flow properties, lower ejection forces, and the best mechanical resistance of compacts. StarLac® showed excellent flow properties, the lowest amounts of undissolved fraction, the highest ejection force values, and the worst compact mechanical resistance. The obtained results revealed that higher compression pressures need to be used or further excipients have to be added to all tested materials in order to improve the friability and tensile strength of formed tablets, except for MicroceLac® 100.

Background: This work is envisaged to understand the use of various superdisintegrants and their drug release effect in the formulation of Ranolazine fast-dissolving tablets (FDTs). Materials and Methods: The FDTs were formulated by using direct compression methods using different superdisintegrants like sodium starch glycolate, croscarmellose sodium and crospovidone. The superdisintegrant's effect at different concentrations was studied with help of precompression studies such as Hausner's ratio, compressibility index, differential scanning calorimetry (DSC) and post-compression studies like friability, disintegration and dissolution studies. Results: The effect of various superdisintegrants on desired drug release was studied and its concentrations were optimized. Based on the evaluation results of various trials, the effective concentration was found to be 15mg/tab of crospovidone. It has a disintegration time of 25 sec and a cumulative percentage drug release of 99.77±0.41. Conclusion: In this study, Ranolazine FDTs were developed and successfully optimized. The ideal superdisintegrant and its concentration were selected. Among the various superdisintegrants, crospovidone was more effective compared to other superdisintegrants. Hence, it is recommended to use crospovidone as an ideal superdisintegrant in the Ranolazine FDTs formulations.

All the standard anti-tubercular drugs, well established as standard therapy, are prefer-entially available in formulations compliant with the young adult population. However, their use in the paediatric and geriatric populations is confronted with issues, such as a high likelihood of incorrect dose administration due to practices like dosage form fracture and splitting. This may lead to drug resistance due to misuse and in-accurate dosage administration, the most dreaded and difficult-to-treat stage of tuberculosis. Poor patient compliance and adherence are major issues with the conventional line of therapy. This burden may be more significant in resource-constrained settings, necessitating the creation of simple formulations that are both geriatric and child-friendly. An extensive literature survey has been conducted in this study using databases of Google Scholar, PubMed, and Research Gate, with a focus on specific research works on oro-dispersible films, tablets, and wafer technology loaded with anti-tuberculosis drugs from 2022 to 2010. Mouth dissolving formulation technology is a very novel approach in the arena of tuberculosis therapy. This may pave the way for future researchers to develop different mouth dissolving for-mulations to treat both pulmonary and extra-tuberculosis. This review paper has summarized all the formulation approaches alongside the present state of the art in tuberculosis therapy using mouth dissolving formulations.
