Fullerene–Oligonucleotide Conjugates: Photoinduced Sequence‐Specific DNA Cleavage

Abstract: The guanine‐specific cleavage of oligonucleotides is achieved with the conjugate 1, which contains a fullerene tethered to a tetradecadeoxyribonucleotide Nu(pNu)n. This conjugate was able to form a duplex with a complementary oligonucleotide, and triplexes with appropriate double‐stranded oligonucleotides. Irradiation of the complexes leads to site‐specfic cleavage of the target sequences—probably through the action of singlet oxygen.

“…[1][2][3][4] Pristine C 60 and its stable nanoscale aggregates in water, known as nano-C 60 , [5] have been widely explored for use in biomedical applications including DNA cleavage, [6] anti-HIV drugs, [7] anticancer therapy, [8] and diagnostic imaging. [9] Cytotoxicity studies suggest that the nano-C 60 aggregates can permeate cell membranes, and even the blood/brain barrier.…”

“…[60]Fullerene derivatives exhibit a range of interesting biological activities including inhibition of HIV enzymes [4][5][6][7][8][9][10][11][12] , photoinduced cytotoxicity via selective cleavage of DNA [13][14][15][16][17] and as a smooth muscle relaxant [18][19][20] . In addition, other interesting reports of fullerene derivatives of biologically significant molecules such as lipids 21 , saccharides 22 , oligonucleotides [23][24] and amino acids have appeared in the literature.…”
