Function of Feline Signaling Lymphocyte Activation Molecule as a Receptor of Canine Distemper Virus

Abstract: ABSTRACT. Morbilliviruses use signaling lymphocyte activation molecule (SLAM) as a receptor for their entry to cells. In this study, a complete gene encoding SLAM of a domestic cat was identified. The identity of feline SLAM with canine one was 73%, and feline SLAM formed the same cluster with those of carnivores. Furthermore, feline cell expressing feline SLAM supported growth of canine distemper virus (CDV) as well as that expressing canine one. These results indicated that feline SLAM can function as a rece… Show more

“…For example, MeV suppresses the immune system by binding to the human SLAM on dendritic cells [50,51]. CDV interacts with monkey, dog, and feline SLAM, but less efficiently with the cells expressing human SLAM [52][53][54]. Since receptors are one of the crucial factors to determine the tissue tropism and host range of a virus, it is important to identify the receptor of FeMV [2,53].…”

“…In particular, K72 and S82 are unique to large and small cats among host mammals. This difference may reduce or eliminate the ability of CDV to naturally infect the Feliformia, although it is not critical for interaction of CDV with cat SLAM in vitro [166]. The H protein of a lion CDV strain was shown to potentially infect a broad range of carnivores with moderate affinity, and unique residues of the lion CDV-H protein play a crucial role in infection [167,168].…”

“…There is a need for adaptation of these viruses to grow in hSLAMF1-Vero cells [ 158 , 159 ]. Host range expansion of CDV with adaptation to SLAMF1 species variants has been reported in carnivores [ 160 , 161 ]. In addition, the ability of wild type variants to CDV to adapt and infect primates has recently been reported [ 162 , 163 , 164 ].…”
