Functional analysis of the circadian clock gene timeless in Tribolium castaneum

Abstract: Circadian rhythms are endogenous oscillations with a period of about 24 h driven by a circadian clock. So far, variable oscillators have been found in insects. To explore the circadian clock of Tribolium castaneum, we cloned the clock gene timeless (Tctimeless). Its open reading frame is 3240 bp in length and consists of 10 exons. Tctimeless is highly expressed in the late pupal stage. Tissue-specific expression analysis in late adult stages revealed high expression of Tctimeless in the head, epidermis, fat bo… Show more

“…The ratio of absorbance A at 260 nm to 280 nm ( A 260 / A 280 ratio) was used to assess the yield and purity of RNA. The RNA was used only when A 260 / A 280 > 1.8, and the RNA integrity was further evaluated using 1% agarose gel electrophoresis (Li et al , ).…”

“…Mortality occurring <5 days after injection was attributed to injection injury rather than to target transcript knockdown. In general, mortality due to injection damage was <10% (Begum, Li, Beeman, & Park, ; Li, Yun, Yu, & Li, ). Individuals from six replications of larval RNAi injections were utilized for single pair mating (average eight pairs per replication).…”

“…purely on photoreception via the compound eyes (Komada et al, 2015;Kutaragi et al, 2018). In other species that lack cry1 (i.e., beetles and cockroaches), tim1 may play a different role in the circadian rhythm generation (Werckenthin et al, 2012(Werckenthin et al, , 2020Li C-J et al, 2018), which stresses the complexity in evolution of divers sets of clock genes in different insects. Recently, a model which includes two different pacemakers with different clock gene sets was proposed for the cockroach to explain per, cry2, and tim1 function in parallel (Werckenthin et al, 2020) (see Figure 1).…”
