Newcastle disease (ND), a highly contagious viral infection, can be transmitted between various bird species, is characterized as high mortality and morbidity and accounted for a great loss in poultry industries worldwide; therefore, the OIE-World Organization for Animal Health has classified it as a notifiable disease (Kristina et al., 2012). To date, tremendous efforts have been put into the investigation on curative treatments including oral administration, eye drop, course spray, intranasal instillation and muscle injection (Meulemans, 1988); nevertheless, the poor efficacies of these therapies lead to ineffective prevention and ND outbreaks. Increasing studies revealed that early protection is a critical process toward successful control of ND transmission (Sharma & Burmester, 1982). In ovo technology was invented and became available for vaccine delivery about 46 years ago (Ricks, Avakian, Bryan, Gildersleeve, & Williams, 1999). In ovo vaccination was used by many modern large scale poultry units as it excelled in inducing early immunity (Sharma & Burmester, 1982). A selective and sensitive liquid chromatography-tandem mass spectrometry method was developed and validated for investigating the pharmacokinetics of umbelliferone, apigenin, genkwanin and hydroxygenkwanin after oral administration of Daphne genkwa extract. Plasma samples were treated by protein precipitation with acetonitrile. Analytes were detected by triple-quadrupole MS/MS with an ESI source in negative selection reaction monitoring mode. The transitions of m/z 161 → 133 for umbelliferone, m/z 269 → 117 for apigenin, m/z 283 → 268 for genkwanin and m/z 299 → 284 for hydroxygenkwanin were confirmed for quantification. Chromatographic separation was conducted using an Eclipse XDB-C column, and the applied isocratic elution program allowed for simultaneous determination of the four analytes for a total run time of 2.5 min. The linearity was validated over the plasma concentration ranges of 1.421-1421 ng/mL for umbelliferone, 0.845-845 ng/mL for apigenin, 1.025-1025 ng/mL for genkwanin and 0.845-845 ng/mL for hydroxygenkwanin. The extraction recovery rate was >82.7% for each analyte. No apparent matrix effect was observed during the bioanalysis. After full validation, the proposed method was successfully applied to compare the pharmacokinetics of these analytes between normal and arthritic rats.

Pidotimod is a synthetic dipeptide with immunomodulatory activity, but the capacity of pidotimod as an immunologic adjuvant to improve vaccine immunogenicity remains unclear. This study evaluated the effectiveness of pidotimod on immune responses of chickens to Newcastle disease (ND) vaccine, and its efficiency to improve protection against highly virulent ND virus (NDV). A total of 300 one-day-old broilers were divided into five groups (numbered I to V). Group I were unvaccinated and served as blank control group. Chickens in Group II to V were immunised with ND vaccine and supplemented with 0, 0.5, 1, 2 g/L of pidotimod in drinking water. The results showed that treatment with pidotimod (1 g/L) remarkably improved (p < .05) the potency of ND vaccine in chickens, as demonstrated by increased immune organ index (organ weight/body weight) and elevated antibody production. Meanwhile, no adverse effect on growth performance was observed. Moreover, as an adjuvant to ND vaccine, pidotimod (1 g/L) also dramatically promoted (p < .05) cell-mediated immunity, as characterised by increased lymphocytes proliferation and elevated ratio of CD4 þ to CD8 þ cells following NDV stimulation. Furthermore, administration of pidotimod (1 g/L) was efficient in inducing effective protection against lethal NDV challenge. Collectively, these data indicate that pidotimod may have the potential to be used as an effective vaccine adjuvant and the immunopotentiating effects deserve further study.
