Functionalized PVA –silk blended nanofibrous mats promote diabetic wound healing via regulation of extracellular matrix and tissue remodelling

Abstract: Chronic cutaneous ulcers, a complex pathophysiological diabetic condition, represent a critical clinical challenge in the current diabetes mellitus pandemic. Consequently, there is a compelling need for bioactive dressings that can trigger healing processes for complete wound repair. Silk fibroin (SF), a natural protein polymer from mulberry and non-mulberry silkworms, has properties that support accelerated wound healing rate. SF from non-mulberry variety possesses additional cell-binding motifs (arginine, gl… Show more

“…However, sufficient vascularized granulation tissue was developed in SF‐treated wounds when compared to untreated. This depicts early recruitment of endothelial cells and angiogenic potential of SF gel owing to the vascularization capacity of both BmSF and AaSF proteins as also reported in previous studies …”

“…BmSF, the mulberry silk variety is known for its wound healing properties and deposition of collagen fibers in various studies . In addition, AaSF possesses Arg cores along with RGD cell binding motifs which aid in faster wound healing owing to its cell‐material interactive properties . Thus, the combination of BmSF and AaSF in SF hydrogel resulted in accelerated healing with normal wound repair process.…”

“…The use of gelatin nanofibers having enough tensile strength for fabricating nonwoven mats has received attention recently for use for antimicrobial applications [45][46][47][48][49]. Successful spinning of minimum diameter nanofibers results in the surface areas of these nanofibers being increased in addition to their light weight.…”

“…Here, the synergistic activity of LL37 and serpin A1 enhanced the nanoparticles antibacterial activity against Staphylococcus aureus ( S. aureus ) and E. coli above the LL37 or serpin A1 alone . More recently, LL37 was functionalized onto PVA/silk fibroin nanofibrous mats by solution blending, and revealed a great efficiency in killing skin bacteria, such as Staphylococcus epidermidis ( S. epidermidis ), and preventing biofilm formation . To the authors' knowledge, functionalization of LL37 has not yet been attempted on the surface of PVA/CA films for prospective applications in wound‐healing.…”
