Fungal Identification Using Molecular Tools: A Primer for the Natural Products Research Community

Abstract: Fungi are morphologically, ecologically, metabolically, and phylogenetically diverse. They are known to produce numerous bioactive molecules, which makes them very useful for natural products researchers in their pursuit of discovering new chemical diversity with agricultural, industrial, and pharmaceutical applications. Despite their importance in natural products chemistry, identification of fungi remains a daunting task for chemists, especially those who do not work with a trained mycologist. The purpose of… Show more

“…Methods for BLAST search in NCBI GenBank and Maximum Likelihood phylogenetic analysis, as utilized to identify fungal strains, have been detailed recently [61]. The BLAST search and phylogenetic analysis using taxon sampling [1] revealed that MSX59553, MSX79542, MSX74391, MSX58124, MSX75296, MSX75281, MSX45374, and MSX70777 belong to the genus Clonostachys (Ascomycota, Hypocreales, Bionectriaceae), while MSX71844 was identified as Purpureocillium lavendulum (Ascomycota, Hypocreales, Ophiocordycipitaceae) (Fig.…”

“…Mycologists have traditionally used morphological characters to identify fungi to the species level but they can often be mislead due to hybridization, cryptic speciation, and convergent evolution [11]. Molecular tools (DNA sequence-based methods) have been useful to solving doubts in the identification process at the species level.…”

“…In all, it has been recommended [96] that additional research of new primers and probes for the characterization of Monilinia species should be conducted to make the method more transferable among qPCR platforms and laboratories [96]. We refer to Raja et al [104] for more information on the merits and demerits of the use of "molecular tools" in fungi detection and identification.…”
