General Method for the Isolation of Plasmid Deoxyribonucleic Acid

Abstract: Plasmid deoxyribonucleic acid (DNA) ranging from 5 x 106 to 65 x 106 daltons may be isolated from chromosomal DNA by the preferential precipitation of the higher-molecular-weight chromosomal DNA in the presence of sodium lauryl sulfate and a high concentration of NaCl.

“…Clone 1 was used for sequencing. Nucleotide sequencing of Daudi 32m cDNA Plasmid DNA was purified by CsCl/ethidium bromide banding from lysozyme/SDS lysates (Guerry et al, 1973). The restriction enzyme sites HinfI, HhaI and HpaII, shown in Figure 1, were used for end labeling and sequencing by the methods of Maxam and Gilbert (1980).…”

“…The isolation procedure was developed from the principles of lysozyme binding in aqueous solution (Metcalf & Diebel 1969) and SDS-salt precipitation (Guerry et al 1973). Bacteria from 40 ml of late log or early stationary phase culture in M17 broth (Terzaghi & Sandine 1975) were harvested, washed twice in ice cold distilled water, using standard centrifugation procedures and resuspended in 36 ml of 25% (w/v) sucrose solution.…”
