Generation and characterization of bioluminescent xenograft mouse models of MLL-related acute leukemias and in vivo evaluation of luciferase-targeting siRNA nanoparticles

Abstract: Chromosomal translocations involving the MLL gene on 11q23 present frequent abnormalities in pediatric, adult and therapy-related acute leukemias, and are generally associated with aggressive disease and poor prognosis. Here, we report bioluminescent acute leukemia xenograft mouse models of the most frequent and aggressive MLL-related acute leukemias (infant and adult MLL-AF9, MLL-ENL, MLL-AF4). Four acute leukemia cell lines carrying MLL-related translocations were stably transduced with a firefly luciferase … Show more

“…MOLM-13 is an adult MLL-AF9 cell line carrying a FLT3-ITD mutated allele that induces very aggressive disease in immunocompromised mice. 63,64 The FLT3-ITD mutation induces constitutive activation of JAK/STAT and Erk1/2 signaling leading to rapid proliferation contributing to the aggressiveness of MOLM-13 cell growth in vivo . Activation of STAT5 dependent signaling is known to promote leukemia stem cell self-renewal.…”

“…Firefly bioluminescence is a two-step reaction (Scheme ): the luciferin molecule reacts with adenosine 5′-triphosphate-Mg 2+ (ATP-Mg 2+ ), thus resulting in the formation of an adenylyl intermediate; the second step is the oxidation of this latter intermediate in the presence of molecular oxygen, generating oxyluciferin, adenosine 5′-monophosphate (AMP), and carbon dioxide. , The generated oxyluciferin is already formed in a singlet excited state, which decays to the ground state with emission of visible light. , Both computational and experimental studies have appointed the keto anion as the light-emitting species. However, no experimental study has ever supported these results in a definitive manner. Useful features of this light-emitting system (as ATP-dependence, high quantum yield, low background noise, high speed of reaction, among others) have triggered the development of numerous applications in pharmaceutical, biomedical, and bioanalytical areas. …”

“…In addition, the potential of BLI has been investigated in a variety of tumor models such as prostate cancer [51], colorectal carcinoma [52], lung cancer [53], pancreatic cancer [54], osteosarcoma [55], glioblastoma [56], and acute lymphoblastic leukemia [57]. Several non-viral methods for in vivo delivery of siRNA have also been investigated and optimized using BLI.…”
