Generation and characterization of PDGFRα‐GFPCreERt2 knock‐In mouse line

Abstract: Platelet-derived growth factor (PDGF) and its receptor play an important role in embryogenesis. PDGF receptor α (PDGFRα) is expressed specifically in the embryonic day 7.5 (E7.5) mesoderm and in the E9.5 neural crest among other tissues. PDGFRα-expressing cells and their descendants are involved in the formation of various tissues. To trace PDGFRα-expressing cells in vivo, we generated a knock-in mouse line that expressed a fusion protein of green fluorescent protein (GFP), Cre recombinase (Cre), and mutated e… Show more

“…60 In the heart, PDGFRα is robustly expressed in fibroblasts, with relatively limited expression in other cell types, including smooth muscle cells 61 and possible expression in cardiac progenitor cells; 62 it has been utilized for the development of an inducible Cre system, the PDGFRα-GFPCreERT2 knock-in mouse line. 63 This will likely prove a useful tool in characterizing CFs and their role in fibrosis. PDGFRβ has also been used as a marker of fibroblasts.…”

“…Furthermore, Platelet-derived growth factor receptor α (Pdgfrα) and platelet-derived growth factor receptor β (Pdgfrβ) belong to the family of receptor tyrosine kinases (42). Pdgfrα and Pdgfrβ play important roles in forming the vasculature during embryonic development and are expressed in cardiovascular and mesenchymal progenitor cells (43,44). Pdgfrβ regulates the development of vascular smooth muscle cells from epicardium-derived cells (45).…”
