Generation, Control, and Processing of Cellular Calcium Signals

Abstract: In the course of evolution, Ca2+ has emerged as the most versatile intracellular messenger. Its concentration within cells is controlled by reversible binding to specific classes of proteins that act as Ca2+ sensors to decode its information before passing it on to targets. The decoding operation is based on specific conformational changes in the sensor proteins. Other proteins intrinsic to membranes simply control Ca2+ concentration without processing its message, by transporting it across membrane boundaries… Show more

“…It has yet to be determined if this represents the function of different androgen-binding proteins with a distinct cell type-dependent distribution or if there is one membrane androgen-binding protein that conveys its signal in a manner subject to other cell type-specific differences. Regardless, by altering [Ca 2+ ] i in unique ways the androgen exposed cell can use the same signaling molecule to specifically regulate different cellular functions [50].…”

“…For example, the Na + / Ca 2+ exchanger has a low affinity, but high capacity, resulting in the transport of up to 5000 ions per second (Carafoli et al, 2001). Such a transporter is useful when Ca 2+ is present in high concentrations (e.g.…”

“…Besides providing energy, mitochondria pump cytosolic Ca 2+ and Mg 2+ , and therefore modulate the cell's [Ca 2+ ] and [Mg 2+ ] (Carafoli et al, 2001). This may be particularly important during calcification when the concentration of these ions increases locally.…”

“…PMCAs extrude Ca 2+ ions from the cell and may thus contribute to the regulation of global, and sub-cellular, concentrations of intracellular free Ca 2+ ([Ca 2+ ] i ) (Karaki et al, 1997;Carafoli et al, 2001;Cartwright et al, 2011). However, it is now evident that PMCAs also have important roles as scaffold proteins whereby they may play a key role in signal transduction (Schuh et al, 2001;Williams et al, 2006;Cartwright et al, 2007;Strehler, 2015).…”
