Generation of autochthonous mouse models of clear cell renal cell carcinoma: mouse models of renal cell carcinoma

Abstract: Renal cell carcinoma (RCC) is one of the 10 most common cancers worldwide, and to date, a strong systemic therapy has not been developed to treat RCC, even with the remarkable modern advances in molecular medicine mostly due to our incomplete understanding of its tumorigenesis. There is a dire unmet need to understand the etiology and progression of RCC, especially the most common subtype, clear cell RCC (ccRCC), and to develop new treatments for RCC. Genetically engineered mouse (GEM) models are able to mimic… Show more

“…Indeed transgenic mice with reporter alleles display specific expression of PAX8 in adult PTC and FTSEC 20,36 . Importantly, VHL inactivation 36 , when combined with other genetic lesions in Pax8-expressing cells, leads to neoplastic lesions of different stage and penetrance 37,38 suggesting proximal tubules as a potential source of RCC.…”

“…“Mouse models” recapitulate physiological variables contributing to disease progression, which are absent in vitro and allow for the derivation of clinically relevant outputs. We describe briefly a few mouse models and assays employed in tumor biology; interested readers may refer to other detailed articles [70,71,72]. The metastatic cascade is routinely captured by spontaneous models for metastasis that involves ectopic/orthotopic tumor cell transplantation in immunocompromised mice.…”

“…In addition, no suitable genetically engineered mouse model (GEMM) for RCC that recapitulates with fidelity the metastatic process exists to date [30][31][32][33] . It is true that around 10 different GEMMs for RCC have been developed 34 37 . The HR reported in their article is lower than in our study.…”
