Genetic and molecular characterization of a distiller’s yeast

Abstract: Auxotrophic mutants have been induced by UV-irradiation and EMS treatment in meiotic segregants of a distiller's production strain of yeast, In complementation tests some of the mutants were allelic to adel, ade3, lys2, met13, trpl, ural and ura4 of standard strains orS. cerevisiae.Crosses were made between two spore-derived clones of the distiller's strain and haploid S. cerevisiae strains carrying several auxotrophic markers. Tetrad analysis of the mating products revealed that the spore-derived clones were … Show more

“…Of the four chromosomes III, three were lacking HML and one HMR. These results indicate that strain A is tetraploid and heterozygous for a number of tested chromosome regions (20). The acquired detailed genetic knowledge about strain A as well as the finding that it contains HO, a gene for homothallism, can now be taken into account in efforts to improve the performance of this strain by using its inherent genetic variation in breeding.…”

“…By tetrad analysis of restriction endonuclease fragment polymorphisms, KEIDING (20) has shown that spore clones of strain A were disomic for at least 7 chromosomes, and that four different chromosomes IlI are present in the strain. Of the four chromosomes III, three were lacking HML and one HMR.…”

“…While the LEU2 allele associated with SalI fragment III is present in industrial S. carlsbergensis and S. cerevisiae strains (15,22,23), the LEU2 allele associated with SalI fragment IV has, until now, only been detected in S. carlsbergensis lager brewing strains, S. monacensis, S. bayanus and S. uvarum (22,23,24).…”

“…Restriction endonuclease fragment analyses by molecular hybridisation with radioactively labelled, cloned gene probes are useful for monitoring gene polymorphisms in Saccharomyces strains (15,18,19,20,21,22,23). In a previous paper, top fermenting ale strains could be distinguished from bottom fermenting (lager) strains by polymorphisms at the HIS4 and the RDN1 loci (20), but only the Bavarian Pschorr Abbreviations: SSC = 0.15 M-NaC1, 15 mM-Na citrate; TBE = 90 mM-Tris base, 90 mM-boric acid, 2 mM-EDTA, pH 8.3; TE = 10 mM-Tris HC1, 1 mM-EDTA, pH 7.5. strain could be separated from other bottom fermenting strains.…”
