Genetic Diversity among Clinical Isolates of Candida glabrata Analyzed by Randomly Amplified Polymorphic DNA and Multilocus Enzyme Electrophoresis Analyses

Abstract: The genetic diversity of 47 clinical and reference strains of Candida glabrata from several geographical origins and diverse clinical disorders, with different antifungal susceptibilities, as well as their genetic relationships were studied through multilocus enzyme electrophoresis (MLEE) and randomly amplified polymorphic DNA (RAPD) techniques. The genetic diversity estimated for 11 MLEE loci measured as average heterozygosity (h) was 0.055. A high level of genetic relatedness among isolates was established b… Show more

“…The yeast microflora isolated from symptomatic women and those from asymptomatic women had similar species and genotypic diversities (Xu et al, 1999, Boldo et al, 2003. However, it was noted from symptomatic women that, a single host can be colonized with multiple species or multiple genotypes of the same species, a trend that is in keeping with other studies Xu et al, (1999), Kam & Xu (2002).…”

“…(1) or absence (0) of a determined band was incorporated in a database, and coefficients of similarity were generated by the bandbased method of Jaccard. 22 A phenogram showing the similarities of the hlyA gene was then generated by the unweighted pair group method with arithmetic mean (UPGMA). Analysis and construction of the phenogram was made using the NTSYSpc2.02j software.…”

“…Molecular typing was performed by RAPD method using the following decamer primers: OPE-18 (5′-GGACTGCAGA-3′) and OPA-18 (5′-AGCTGACCGT-3′) (Gibco BRL, USA). Amplification reactions were performed as previously described [4]. To ensure the reproducibility of these polymerase chain reaction (PCR)-based fingerprints, the samples were amplified in three separate RAPD reactions.…”
