Paris polyphylla is a threatened medicinal plant with numerous pharmacological effects including prized anticancer phytochemicals. In the present study, the genetic diversity, and population structure of 32 P. polyphylla germplasm collected from natural habitats of three ecological regions Central, Eastern, and Western Nepal was performed using 15 simple sequence repeat (SSR) markers. The polymorphic information content (PIC) value of the SSR marker ranged from 0 to 0.75. The value of the average observed heterozygosity (Ho = 0.63) is greater than the expected heterozygosity (He = 0.53) with average total heterozygosity (Ht = 0.32). The molecular variance analysis (AMOVA) revealed high genetic variation within the population (74%) than low genetic variance (26%) among populations. The value of Fst and gene flow were 0.53 and 0.458, respectively. In STRUCTURE analysis 32 germplasm were separated into two populations where Eastern accessions were separated far from Central and western accessions which were also in complete correspondence to the unweighted pair group method based on arithmetic average (UPGMA) phylogenetic tree and principle coordinate (PCoA) analysis. Further, in the UPGMA and PCoA, accessions collected from the same or relatively similar geographic origin were closer. These genetic results are of great significance for scientifically formulating conservation strategies and commercial cultivation for the medicinally important Vernalable plant P. polyphylla.

Epigenetic and molecular variation is a key approach for the improvement of plants. From in vitro tissue culture of Dendrocalamus farinosus; 30 plants were regenerated and after 2 years, the physiological, biochemical, and genomic studies were conducted. The results highlighted that for all phenotypic characteristics, the 30 regenerated plants were superior in comparison with the control (CK). From genetical analysis, a total of 97 bands were witnessed ranging between 212 bp and 2.2 kb. The results for OPU14 showed one additional specific band (723 bp) and one band (700 bp) were missing. The 10 plants were having genetic variability and can be termed as somaclones while the other plants have epigenetic variations. The cellulose and lignin analysis highlighted that somaclone No. 30 has the least cellulose content of 35%; while the somaclones No. 102 and No. 213 have the least 3.21% and 3.48% of lignin contents. Therefore, somaclones No. 30, No. 102, and No. 213 were selected for the histochemical localization. The lignin investigation revealed that somaclone No. 30 is greater while somaclones No. 102 and No. 213 were reduced in vascular bundles in comparison with the CK along with the high expression level of 4CL, C3H, C4H, COMT, and CCoAOMT1 genes.

Nepenthes adrianii is one of pitcher plant species that grows endemically in Mount Slamet, Central Java. At present, it is one of pitcher plant collections of Baturraden Botanical Garden. Since N. adrianii is dioecious and both sexes are difficult to distinguish morphologically, early sex determination supporting its conservation at Baturraden Botanical Garden is needed. One approach can be performed with the use of RAPD molecular markers. Hence, this study aims to know whether differences in RAPD pattern between male and female N. adrianii exist or not and also to find out what the differences are. Genomic DNAs were extracted from leaves of 4 males, 2 females, and 2 sexually unidentified individuals. The extracted DNAs were then used to analyze DNA variation between male and female N. adrianii employing RAPD technique. As many as five oligonucleotide primers (OPA-15, OPK-16, OPP-15, OPP-08, and OPO-08) were used to amplify N. adrianii DNA. The results showed that one primer, i.e. OPK-16 (5’-GAGCGTCGAA-3’), produces a specific band of approximately 290 bp which is only found in female plants. It is assumed that this band is related to gene(s) controlling sex determination in N. adrianii. The RAPD marker can be used for the sex determination of young N. adrianii seedlings.
