Genetic heterogeneity of L-Zagreb mumps virus vaccine strain

Abstract: Background: The most often used mumps vaccine strains Jeryl Lynn (JL), RIT4385, Urabe-AM9, L-Zagreb and L-3 differ in immunogenicity and reactogenicity. Previous analyses showed that JL, Urabe-AM9 and L-3 are genetically heterogeneous.

“…cDNA was prepared from total RNA using random hexamers and MuLV reverse transcriptase as reported previously (Košutić -Gulija et al, 2008). Nested PCRs were performed for HN, F and P genes.…”

“…Further studies based on reverse genetics are required to investigate the relationship between these diversities and the molecular mechanism of attenuation and/or virulence. In spite of the fact that genetic heterogeneity in other mumps vaccine strains has also been observed (Boriskin et al, 1992;Amexis et al, 2002;Sauder et al, 2006;Kosutic-Gulija et al, 2008;Tillieux et al, 2009), it might be necessary to prepare a cloned attenuated vaccine RS-12 virus as a genetically homogenous seed. There is currently limited data available on sequencing of the full-length genomes of attenuated MuV vaccine strains.…”

“…L-Zagreb variants A and B were plaque purified and their genomes were completely sequenced [13]. Genomes of the two variants differ in nt G1073T, C5261T and G11345T, the first letter indicating the nt in variant A cDNA and the second in variant B cDNA, respectively.…”

“…Sequences spanning nt site 5261 from both plaque purified viral variants were PCR amplified and inserted into pUC19 plasmid (New England Biolabs, USA), thus obtaining plasmid variants A and B (PVA and PVB, respectively) [13]. PVA had a cytidine (C) at position 5261 of the mumps cDNA and PVB had a thymidine (T).…”
