Genetic manipulation of 6-phosphofructokinase in potato tubers

Abstract: Abstract. The aim of this work was to discover whether genetic manipulation of 6-phosphofructokinase [EC 2.7.1.11; PFK(ATP)] influenced the rate of respiration of tuber tissue of Solanum tuberosum L. Transgenic plants were produced that contained the coding sequence of the Escherichia coli pJkA gene linked to a patatin promoter. Expression of this chimaeric gene in tubers resulted in a 14-to 21-fold increase in the maximum catalytic activity of PFK(ATP) without affecting the activities of the other glycolytic … Show more

“…These measurements were confirmed by recovery experiments, have low standard errors, and the wild-type values are comparable to those previously reported for potato tubers (e.g. Jelitto et al, 1992;Burrell et al, 1994). In U-IN2-30 there is a significant three-to fourfold increase in the hexose phosphate pools relative to the wild-type.…”

“…The levels of 3-phosphoglycerate (3PGA), phosphoenolpyruvate (PEP) and pyruvate were comparable to those measured previously in potato tubers (e.g. Burrell et al, 1994;Jellito et al, 1992), and the measurements were authenticated by recovery experiments. The measurements were made on the same extracts that were used for the hexose phosphate measurements.…”

“…The contents of Glc, Fru, Suc, starch, and adenylates were determined as in Mustroph et al (2006a) and phosphorylated metabolites as in Burrell et al, (1994). The content of Pi was determined as described by Itaya and Ui (1966).…”
