Genetic response to an environmental pathogenic agent: HLA‐DQ and onchocerciasis in northwestern Ecuador

Abstract: The aim of this study is to explore human leukocyte antigen (HLA)-DQ variability in two populations (Cayapas Amerindians and Afro-Ecuadorians) who live near one another along the Cayapa River and who are exposed to the same environmental stresses, such as infection by Onchocerca volvulus. HLA-DQA1 and HLA-DQB1 of 149 unrelated individuals (74 Cayapas and 75 Afro-Ecuadorians) have been analyzed. HLA high-resolution molecular typing was performed by sequence-based typing, sequence-specific oligonucleotides hybri… Show more

“…The choice for these regions was due to their crucial role in encoding the HLA molecules; in fact, these exons, respectively, encode for the α 1 and β 1 regions of the HLA‐DQ subunits, which are the regions that form the antigen cleft. The primers for amplification and sequencing are listed in Table S1; they were used according to references (Cordovado et al ., ; van Dijk et al ., ; De Angelis et al ., ). Conditions for the polymerase chain reaction (PCR) assay by custom‐designed primers were as follows: 10 min at 95°C, 35 cycles of 30 s at 95°C, 1 min at 53.5°C, 30 s at 72°C and a final extension of 7 min at 72°C.…”

“…This research aimed to analyse the HLA‐DQ profile of this population to ascertain its genetic relationships with neighbour groups. Several studies of HLA class I and class II variation in Amerindian populations have been carried out for anthropological as well as evolutionary and clinical studies (Tsuneto et al ., ; Parolin & Carnese, ; Arnaiz‐Villena et al ., ; Vargas‐Alarcon et al ., ; De Angelis et al ., ). These data were compared with other Amerindian groups and worldwide populations to clarify the relationships among northern inhabitants of South America and to highlight the power of HLA system in determining population relationships.…”

“…Each donor was asked to supply name, birthplace, language and ethnicity for three generations, in order to allow us to determine the extent of the recent admixture. Further information about these human groups and DNA purification procedures is available in previous studies (De Stefano et al, ; Polimanti et al, ; De Angelis et al, ). We matched our data with those reported for different ethnic groups in the HapMap project (available at http://hapmap.ncbi.nlm.nih.gov) (International HapMap3 Consortium et al, ).…”

“…However, the clinical features of the Yemeni form of onchocerciasis are different from that described in Africa. It is noteworthy that a possible role of host genetics could not be ruled out as a determinant of the clinical profile of onchocerciasis by determining the nature of host immune response against the parasite (De Angelis et al, 2012;Donfack et al, 1999;Meyer et al, 1994).…”
