Genome organization and phylogenetic relationship of Pineapple mealybug wilt associated virus-3 with family Closteroviridae members

Abstract: The nucleotide sequence of Pineapple mealybug wilt associated virus-3 (PMWaV-3) (Closteroviridae: Ampelovirus), spanning seven open reading frames (ORFs) and the untranslatable region of the 3' end was determined. Based on the amino acid identities with orthologous ORFs of PMWaV-1 (54%-73%) and PMWaV-2 (13%-35%), we propose PMWaV-3 is a new species in the PMWaV complex. PMWaV-3 lacks an intergenic region between ORF1b and ORF2, encodes a relatively small, 28.8 kDa, coat protein, and lacks a coat protein duplic… Show more

“…With BYV, CTV and GLRaV-2 (Besse et al, 2009) CPm coats an extremity of the virions, forming a distinct structure for which the terms 'rattlesnake' or 'heterodimeric' were coined. This structure may not be a general characteristic of the family, as suggested by Agranovsky et al (1995) based on the information available at that time, because the ampelovirus species of subgroup II lack a CPm gene (Melzer et al, 2008;Sether et al, 2009; …”

“…The isolate of PMWaV-1 detected in this study is similar to that reported in Cuba (Figure 2) due perhaps to the highest trade exchange of propagative material between the two countries and least similar to isolates from Australia and Hawaii related to differences in systems production, cultivated plant material and prevailing environmental conditions in all three cases (Sether et al, 2010). Like PMWaV-1, PMWaV-3 does not induce MWP by itself or together with the mealybug feeding in Hawaii (Sether et al, 2005) and it is unknown what is its effect on the production of pineapple (Sether et al, 2009). Only one plant from the same location that showed different symptoms resulted positive for PMWaV-3.…”

“…Así como el PMWaV-1, PMWaV-3 no induce la MaPHP por sí solo ni junto con la alimentación del de piojo harinoso en Hawai (Sether et al, 2005) y no se sabe cuál es el efecto en la producción de piña (Sether et al, 2009 vector management and water supply is neglected since it has been shown that infection by this virus and limited irrigation produce a negative effect (6.7 and 4.2 %, respectively) and an additive effect (13.4 %) on size and weight of the pineapple fruit .
