Genome-wide association analysis identifies genetic correlates of immune infiltrates in solid tumors

Abstract: Therapeutic options for the treatment of an increasing variety of cancers have been expanded by the introduction of a new class of drugs, commonly referred to as checkpoint blocking agents, that target the host immune system to positively modulate anti-tumor immune response. Although efficacy of these agents has been linked to a pre-existing level of tumor immune infiltrate, it remains unclear why some patients exhibit deep and durable responses to these agents while others do not benefit. To examine the influ… Show more

“…Additionally, KRAS mutations were found to be significantly associated with lower levels of B cells, a relationship that has not been described previously. CDKN2A mutations were found to be associated with lower infiltration of CD8 + Tregs, which is in line with previous findings of lower levels of immune cell infiltration in a wide range of solid tumors with CDKN2A mutation, including pancreatic cancer . Mutations in SMAD4 and APC were found to be associated with higher abundance of several CD8 + lymphocyte subsets and in the case of APC also with CD4 + lymphocytes.…”

“…43,44,60 Interestingly, miRNA-375 is strongly associated with the Wnt/ b-catenine pathway 49 and the Hippo pathway, 59,[61][62][63] known to be involved in ICIs resistance. [64][65][66][67][68][69][70] Moreover, miRNA-375 targets the JAK2 gene, 62,71 involved in the IFN-g and TNF-a pathways, key-regulators of cytotoxic CD8C immune response. 72 Taken together, these data, highlighted by our results, suggest an implication of both miRNA-320b and miRNA-375 in anti-tumour immune response with ICIs, and the potential utility of screening for these miRNA in plasma before beginning nivolumab to select patients who will have response and clinical benefit with this treatment.…”
