Genomic Sequence of Klebsiella pneumoniae IIEMP-3, a Vitamin B 12 -Producing Strain from Indonesian Tempeh

Abstract: Klebsiella pneumoniae strain IIEMP-3, isolated from Indonesian tempeh, is a vitamin B12-producing strain that exhibited a different genetic profile from pathogenic isolates. Here we report the draft genome sequence of strain IIEMP-3, which may provide insights on the nature of fermentation, nutrition, and immunological function of Indonesian tempeh.

“…Vitamin B12 in tempeh was naturally produced by bacteria, for example, K. pneumoniae and C. freundii (Areekul et al., 1990; Keuth & Bisping, 1993; Liem et al., 1977; Okada, 1989). The vitamin B12‐producing K. pneumoniae strains in tempeh were not pathogenic given that they did not produce enterotoxin and had different genetic profiles compared to those pathogenic to humans (Keuth & Bisping, 1993; Yulandi et al., 2016). In Indonesia, harmless Klebsiella spp.…”

“…Some K. pneumoniae isolates from EMP and WJB tempeh fully sequenced and subjected to genomic comparison to pathogenic strain in the previous study [29]. K. pneumoniae is known to produce vitamin B 12 during tempeh fermentation [30]. Data analysis from the amplicon sequencing of metagenomic samples of EMP and WJB tempeh in the previous study [4,5] failed to reveal this important genus.…”
