Genomics of Helper Component Proteinase Reveals Effective Strategy for Papaya Ringspot Virus Resistance

Abstract: Papaya ringspot virus (PRSV) causes severe economic losses in both cucurbits and papaya throughout the tropics and subtropics. Development of PRSV-resistant transgenic plants faces a major hurdle in achieving resistance against geographically distinct isolates. One of the major reasons of failing to achieve the broad-spectrum PRSV resistance is the involvement of silencing suppressor proteins of viral origin. Here, based on sequence profile of silencing suppressor protein, HcPro, we show that PRSV-HcPro, acts … Show more

“…One gram of leaf tissues from PRSV-infected plants was ground in 0.1 M potassium phosphate buffer, pH 7.0. Plants were dusted with Carborundum and PRSV-infected leaf extract was inoculated on the youngest fully-expanded leaves of each plant as described by Mangrauthia and coworkers [30].…”

“…PRSV infection was detected and quantified serologically using the plate trapped-antigen enzyme-linked immunosorbant assays (ELISA) method [30] in Maxisorb microtiter plates (Nunc, Roskilde, Denmark), using 0.1 g fresh leaf tissue. All incubations were performed at room temperature, unless stated otherwise.…”

“…In planta expression of PRSV-HcPro affects developmental biology of plants, suggesting the interference of suppressor protein in micro RNA-directed regulatory pathways of plants. Besides facilitating the establishment of PRSV, it showed strong positive synergism with other heterologous viruses as well [97].…”

“…In planta expression of PRSV-HcPro affects developmental biology of plants, suggesting the interference of suppressor protein in micro RNA-directed regulatory pathways of plants. Besides facilitating the establishment of PRSV, it showed strong positive synergism with other heterologous viruses as well (Mangrauthia et al, 2010). Therefore, resistance in transgenic papaya can be overcome by PRSV with distant homology to the transgene, or by PRSV strains with HCPro that can sufficiently suppress the silencing mechanism of transgenic papaya.…”
