Abstract: Es wird ein Literaturüberblick zur Gewinnung und Charakteristik von Proteasen aus thermophilen Mikroorganismen gegeben. Nach einem allgemeinen Überblick zur Problematik der Thermophilie wird auf die von Schimmelpilzen und Bakterien produzierten proteolytischen Enzyme eingegangen. Besonders hervorgehoben werden die Protease aus Streptomyces rectus var. proteolyticus, die „Aminopeptidase I”︁ aus Bacillus stearothermophilus sowie das Thermolysin aus B. thermoproteolyticus.

A brief review of the most important properties and fields of application of microbial proteinases are presented. Special possibilities of an industrial utilization of these enzymes are discussed: in the photographic industry for the production and degradation of gelatin as well as for silverless photography, in the food industry for the improvement of the functional properties of proteins as well as for increasing their nutritive value, in organic synthesis for the production of peptides, and in sewage treatment plants for the stabilization of sludge.

A dried proteolytic enzyme preparation “Thermitase” is produced of the cultural solution of Thermoactinomyces vulgaris. The procedure comprises concentration, sedimentation, ethanol and sodiumsulfate precipitation and drying in laboratorial and semitechnical scale. The elimination of rape seed oil which impairs the salt sedimentation was specially taken into account. A certain sedimentation procedure using caoline or a aqueous solution of CaCl2 and Na2HPO4 or filtration was elaborated to eliminate the lipid components in the concentrated solution. The method is convenient to get good yields of the thermostable protease.

Nach einem kurzen historischen Rückblick auf die Nutzung enzymologischer Wirkprinzipien bei der Lebensmittelproduktion und den zunehmenden Einfluß der Enzymforschung auf eine wissenschaftliche Durchdringung der hierbei ablaufenden Mechanismen werden Stand und Trends der Enzymapplikation anhand einiger ausgewählter Beispiele aufgezeichnet sowie die derzeitig wichtigsten auf diesem Gebiet durchgeführten Entwicklungsarbeiten erörtert. Hierbei wird in 3 Gruppen (lösliche Enzyme, immobilisierte Enzyme und aromabildende Enzyme) unterteilt. Im Vordergrund der Ausführungen steht der Nahrungsgütersektor. Darüber hinaus werden einige Fragen diskutiert, die für die Zukunft von allgemeinerem Interesse sein dürften.
