Glucose-Responsive Vehicles Containing Phenylborate Ester for Controlled Insulin Release at Neutral pH

Abstract: This study is devoted to developing amphiphilic block polymers based on phenylborate ester, which can self-assemble to form nanoparticles, as a glucose-sensitive drug carrier. Poly(ethylene glycol)-block-poly[(2-phenylboronic esters-1,3-dioxane-5-ethyl) methylacrylate] (MPEG5000-block-PBDEMA) was fabricated with MPEG5000-Br as a macroinitiator via atom transfer radical polymerization (ATRP). Using the solvent evaporation method, these block polymers can disperse in aqueous milieu to self-assemble into micellar… Show more

“…[22][23][24][25][26] The covalent interactions between phenylbronic acid and the hydroxyl groups of saccharides (1,2-or 1,3-diols) lead to the formation of five-or sixmembered rings and allow bronic acid to be a sensitive detector for saccharides. [25][26][27][28] Figure 4 shows the fluorescent intensity curves vs. glucose concentration controlled at 0-0.8 mg/mL and the concentration of polymer kept at 0.5 wt%. The fluorescent intensity is decreased with the increasing glucose concentration in the solution.…”

“…The percentage release values observed and the release rates are broadly in agreement with the literature. [49][50][51] of these two materials (ie, had a lower LCST) and further, since AAPBA is very expensive, is more cost-effective than PAD-2-1. PAD-5-1 was thus taken forward into further work.…”

“…According to the reported method, [ 35 ] FITCinsulin could be loaded to the Y-shaped polymer nanoparticles with interactions of hydrogen bonding, van der Waals force and the hydrophilic-hydrophobic interaction. Through multiple ultrafi ltration centrifuge (MWCO 30 kDa) to completely remove free FITC-insulin, the insulin-loading particles were further measured by DLS and zeta potential.…”
