Glycolysis and glutaminolysis cooperatively control T cell function by limiting metabolite supply to N-glycosylation

Abstract: Rapidly proliferating cells switch from oxidative phosphorylation to aerobic glycolysis plus glutaminolysis, markedly increasing glucose and glutamine catabolism. Although Otto Warburg first described aerobic glycolysis in cancer cells >90 years ago, the primary purpose of this metabolic switch remains controversial. The hexosamine biosynthetic pathway requires glucose and glutamine for de novo synthesis of UDP-GlcNAc, a sugar-nucleotide that inhibits receptor endocytosis and signaling by promoting N-acetylglu… Show more

“…Glycolysis is crucial as energy resource for ocular tissues. However, it can maintain the balance between proinflammatory and anti-inflammatory cytokines (Araujo et al 2017). De novo purine biosynthesis is found to be common pathway when we compare the proteins from SDS-PAGE and 2D electrophoresis.…”

Abundance of hydrolase class of enzymes in vitreous fluid samples may indicate swelling and degeneration of cortical fibres during autoimmune uveitis (Gorthy 1978).…”

“…This was required not only for clonal expansion of peripheral T cells, but also for T-cell progenitor expansion and their malignant transformation into T cell acute lymphoblastic leukemia (T-ALL) (Swamy et al, 2016). Hexosamine biosynthesis may play a critical role in the balance of Th17 and Treg cells, as increased hexosamines promote Treg in a manner similar to that observed in M2 macrophages (Araujo et al, 2017). …”

“…Metabolic flux through glycolysis vs the HBP is competitive and can be gated based on environmental conditions, impacting such functions as cell fate during T‐cell maturation . The HBP deviates from glycolysis as a result of the enzyme glutamine fructose‐6‐phosphate aminotransferase (GFAT) (Figure ).…”
