GnRH Regulation of Jun and Atf3 Requires Calcium, Calcineurin, and NFAT

Abstract: GnRH binds to its receptor on gonadotropes and activates multiple members of the MAPK signaling family that in turn regulates the expression of several immediate early genes (IEGs) including Jun, Fos, Atf3, and Egr1. These IEGs confer hormonal responsiveness to gonadotrope-specific genes including Gnrhr, Cga, Fshb, and Lhb. In this study we tested the hypothesis that GnRH specifically regulates the accumulation of Jun and Atf3 mRNA through a pathway that includes intracellular Ca²⁺, calcineurin, and nuclear fa… Show more

“…Indeed, we also observed increased c-FOS and c-JUN mRNA and protein levels following treatment of immortalized EVT cells with GnRH. These findings are in agreement with earlier studies demonstrating GnRH-induced expression of c-FOS and c-JUN in pituitary cells (9,10). Interestingly, GnRH-induced a more prolonged up-regulation of c-JUN mRNA levels than c-FOS in HTR-8/SVneo cells.…”

“…For example, activator protein-1 (AP-1) is a critical mediator of GnRH-induced gonadotropin expression in pituitary cells (9,10). Both major components of the AP-1 transcription factor, c-FOS and c-JUN, are detected in invasive EVT cell populations (11,12).…”

“…GnRH stimulates activation of MAPK1/3, JNK, and p38 MAPKs through the PKC pathway in gonadotropes (1). Activation of JNK and MAPK1/3 trigger activation of cap-dependent translation and increased expression of IEG's such as Egr1, Atf3, c-Jun, and c-Fos, which regulate both Lhb and Fshb promoters (12,46). Our results in Figures 3 and 4 suggest that activation of MAPK 1/3, Egr1, and Atf3 are more sensitive to ROS inhibition than JNK, c-Fos and c-Jun, but both Lhb and Fshb gene expression require ROS signaling.…”

“…In gonadotropes, GnRH-activated MAP kinases translocate to the nucleus to activate factors that increase Lhb and Fshb transcription, such as the immediate-early response genes c-Fos, c-Jun, Egr1 and Atf3 (1,4,12). c-FOS and c-JUN proteins heterodimerize to form the activator protein 1(AP1) transcription factor, which induces the transcription of the Fshb gene (13).…”

“…The molecular identification of key components in the ethylene signal transduction pathway has been carried out in the model plant A. thaliana, and the complicated regulation of ethylene response and signaling has been comprehensively investigated (Guo and Ecker 2004;Kendrick and Chang 2008;Binder et al 2012). Most of the findings are largely based on the mutagenesis and genetic screening of ethylene insensitive mutants in Arabidopsis.…”
