Gold Glyconanoparticles as Water-Soluble Polyvalent Models To Study Carbohydrate Interactions

Abstract: Glycosphingolipid clustering and interactions at the cell membrane can be modeled by gold glyconanoparticles prepared with biologically significant oligosaccharides. Such water‐soluble gold glyconanoparticles with highly polyvalent carbohydrate displays (see picture, gray hemisphere: gold nanoparticle) have been obtained by a simple and versatile strategy.

“…Most of these new properties arise from the so-called ''size effect'' which affects the electronic structure, as well as from the increase of the ratio of atoms located at the surface with respect to the total number of atoms of the NP [1,2]. Monodispersed gold particles with an average size below 2 nm have been synthesized by different routes using ligands and surfactants as protective agents [3][4][5].…”

“…12,13 Multivalent glycoconjugates based on gold glyconanoparticles (GNP) which allow multiple interactions between their sugars and cell surface receptors 14,15 represent an appealing vaccine delivery technology. Major GNP advantages are their easy handling, resistance to enzymatic degradation, water solubility, convenient antigen loading, immune cell targeting, lack of toxicity and small size around 2 nanometers.…”

“…Gold NPs coated with glycan monolayers have been studied extensively in various areas of biomedical research (Marradi, Martín-Lomas, Penadés, 2010;Chang-Ming, 2011;Reichardt, Martín-Lomas, Penadés, 2013). Since the first report in 2001 (De La Fuente et al, 2001), gold glyconanoparticles (GNPs) received great attention among glycochemists.…”

“…The pioneering group of Penadés has been the first to identify gold NPs as potential scaffolds for vaccine preparation (De La Fuente et al, 2001). Accordingly, in an early report, hybrid glycoNPs have been synthesized by combining the Lewis Y (25) and the TF (26) oligosaccharides tumor markers, a peptide from tetanus toxoid (TT) as T-cell helper (27) FIGURE 10 -Multicomponent TACA vaccine incorporating of the requisite elements to raise both humoral and cytotoxic immunity (Galan, Dumy, Renaudet, 2013).…”
