The cytotoxic and genotoxic effects of goniothalamin, a plant styryllactone, were evaluated using the 3-(4,5 -dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay and the Alkaline Comet assay respectively in human leukemic cell lines. Following 72 h of treatment, the IC 50 values of goniothalamin in human HL-60 promyelocytic leukemia cells and CEM-SS T-lymphoblastic cells were 4.5 µg/mL and 2.4 µg/mL respectively. The genotoxicity of goniothalamin in both HL-60 and CEM-SS cells was detected as early as 2 h following treatment at IC 10 and IC 25 concentrations. However, pretreatment with the antioxidant N-acetyl-cysteine (NAC) at 1 mM for 30 minutes did not abrogate genotoxicity of this compound. This result suggests that primary induction of DNA damage by goniothalamin may not involve oxidative damage. In conclusion, our results demonstrate genotoxic damage induced by goniothalamin in leukemic cells. Further studies are needed to ascertain the mode of action of goniothalamin in inducing DNA damage.

Goniothalamus (Annonaceae) is well known for the rich of styryllactone contents and these plants are famous for the potential to be applied as alternative biological activities. The chemical and biological investigations of Goniothalamus tapis is tested for anticancer activities. The research was processioned to extract, isolate, purify and elucidate structure from the leaves and twigs of G. tapis. The isolated compound was evaluated with cytotoxic activity. Goniothalamin was isolated from ethyl acetate extract of G. tapis and the spectroscopic techniques were used for structure elucidation. In addition, goniothalamin was the most powerful to cytotoxic activity which was the first reported for this specie. The goniothalamin was possessed a potent cytotoxicity. Therefore, it is possible to use this compound as a pharmacological agent.

Many proteins are transported from the nucleus to the cytoplasm by the exportin CRM1, which recognizes cargo proteins through a leucine rich nuclear export signal (NES). This nuclear export process can be inhibited by several small molecules, both natural products and fully synthetic compounds. The structural basis for the inhibition of nuclear export by leptomycin (LMB) based on disruption of the protein/protein interaction between CRM1 and cargo proteins is discussed. The chemistry and inhibition of nucleocytoplasmic transport of leptomycin, anguinomycin and derivatives, goniothalamin, JBIR-02, valtrate, dihydrovaltrate, ACA, peumusolide A and several synthetic compounds is presented. Consequences for the design of nuclear export inhibitors is discussed, and the potential of these compounds as anticancer agents is evaluated.
