Graduate Student Literature Review: Understanding the genetic mechanisms underlying mastitis

Abstract: Mastitis is the costliest disease facing dairy producers today; consequently, it has been the subject of substantial research focus. Efforts have evolved from an initial focus on understanding the etiology of intramammary infections to the application of preventative measures, including attempts to breed cows that are resistant to infection. However, breeding for resistance to infection has proven difficult, given the complexity of the disease and the high expense associated with assembling highquality genotyp… Show more

“…Bovine mastitis is a tricky problem in dairy farming and leads to the decline of milk quality and remarkable economic losses worldwide [ 1 ]. Pathogen invasion to mammary gland is the main cause of this complex disease [ 2 ]. Several microorganisms, including Escherichia coli and Staphylococcus aureus ( S. aureus ), induce bovine mastitis.…”

“…Mediator and inflammatory cytokine expression are activated by Toll-like receptors (TLRs) which are implicated in cell differentiation, apoptosis, and the immune response. Additionally, the immune response and cytokine’s role in the spread of mastitis has been reported by several literatures [ 9 ]. The pro-inflammatory cytokines increase locally and systemically in E .…”

“…Similarly, the SNP of gene CD46 was also found to be associated with mastitis in bovines (93). SNP studies of TGP genes can pave the knowledge discovery of pathway genes mediating mastitis disease by integrative analysis with transcriptomic data (24,94).…”

“…This ensures that minor novel alleles are not missed, which are not present in the prefabricated SNP array/chip (21,22). TGP approach offers multifold advantages like sequencing at higher depth (500-1000X coverage) than the whole genome/exome sequencing, allowing the discovery of extremely rare variants which are potentially the most valuable alleles for association studies (23,24). It is rapid and cost effective, especially in the case of association studies where the sample size is limited and mining of causative mutations in a single assay is imperative (25).…”
