Poly(ethylene glycol) (PEG), a kind of “green” medium, was used to extract artemisinin from the Chinese herb Artemisia annua L. to replace the traditional volatile and explosive solvents like petroleum ether, diethyl ether, etc. PEGs, with different molecular weights, show excellent ability for artemisinin extraction, and the extraction efficiency decreased with the increase of molecular weight. It was also demonstrated that ultrasonic irradiation could further enhance extraction efficiency compared with conventional water bath heating. Results indicate that the extraction amount of artemisinin from one gram of the leaves could reach 15.8 mg/g under the optimal conditions with ultrasonic power of 100 W, the temperature of 50 °C, time of 1 h in PEG-200, which were optimized by response surface method. Compared with Soxhlet extraction using petroleum ether (PE), the method developed here shows higher extraction efficiency (15.8 mg/g for PEG vs. 14.9 mg/g for PE process) and shorter extraction time (1 h vs. ca. 12 h). Additionally, artemisinin in the PEG solution can be recovered using resin adsorber. The results show that the recovery of artemisinin could reach almost 100% by D101 or H103 porous resin, and then the drug could be desorbed from the resin by ethanol or ethyl acetate. Moreover, the kinetic and mechanism studies of the extraction process revealed that ultrasonic irradiation could enhance mass transfer and the hydrogen bonding interactions between artemisinin and PEG further facilitate the separation of artemisinin from the leaves. This work provides an alternative process for efficient artemisinin extraction by using an environmentally benign medium with outfield intensification, which could be potentially extended to the production of other natural drugs.

Curcumin Eutectics with Enhanced Dissolution Rates: Binary Phase Diagrams, Characterization, and Dissolution Studies

Curcumin is a potentially viable pharmaceutical ingredient obtained from the rhizome of a turmeric plant, Curcuma longa. It is a polyphenolic compound which is known to possess antibacterial, anti-inflammatory, antitumor, and anticancer properties. Its use in pharmaceutical applications has been limited because of its poor aqueous solubility and hence poor bioavailability. In this work, attempts were made to formulate new solid forms of Curcumin with several coformers, mainly to enhance the dissolution rate of curcumin in aqueous medium. Ibuprofen, succinic acid, paracetamol, carbamazepine, ethyl paraben, glycine, tyrosine, N-acetyl D,Ltryptophan and biotin are the coformers investigated in this study. Binary phase diagrams were constructed for each binary system which helped in identifying the nature and the composition of the solid phase. All binary systems except curcumin−ibuprofen exhibited eutectic formation. The curcumin−ibuprofen system resulted in a physical mixture. These solid phases were further characterized through powder X-ray diffraction, differential scanning calorimetry, Fourier transform infrared spectroscopy and Raman spectroscopy. Dissolution studies conducted for eutectics showed enhanced dissolution rates as compared to raw curcumin. The results obtained were compared with the literature reports to present a consolidated account of research being conducted to enhance aqueous solubility of curcumin by developing new solid forms of curcumin such as eutectics, coamorphous solids, and cocrystals. Further, attempts have been made to understand how molecular geometry and intermolecular interactions influence the formation of a specific solid form.

Comparative Evaluation of Different Extraction Techniques for Separation of Artemisinin from Sweet Wormwood (Artemisia annua L.)

Sweet wormwood (Artemisia annua L.) valorization is gaining importance due to the presence of the health-promoting bioactive compound, artemisinin. Considering the wide possible application of artemisinin drug formulations, new, greener technologies in their production are welcome. In this study, artemisinin was extracted from A. annua leaves using green extraction technologies (ultrasound-assisted extraction, supercritical CO2 extraction, deep eutectic solvent extraction and subcritical water extraction) in combination with green solvents. Artemisinin was present up to 3.21 µg/mgdw. Among the different green extraction techniques, HPLC data revealed supercritical CO2 (SCO2) extracts to exhibit the highest yield of artemisinin due to the solvent non-polar properties. Additionally, the volatile compounds profile of SCO2 extract was determined, with camphor (12.23%), arteannuin b (15.29%) and artemisia ketone (10.97%) as the most abundant compounds. Obtained results encourage the use of green extraction techniques for the separation of artemisinin and are expected to find potential in pharmaceutical, cosmetic and food applications.
