Growth factor regulation of bovine satellite cell growth in vitro.

Abstract: This study examined the effects of basic fibroblast growth factor (bFGF), insulin-like growth factor I (IGF-I) and transforming growth factor beta (TGF-beta) on the proliferation and differentiation of primary bovine satellite cells (BSC) in vitro. Individually, these three factors had the following effects on satellite cells: bFGF stimulated proliferation (P less than .01) but inhibited differentiation (P less than .05); IGF-I had no effect on proliferation but stimulated differentiation (P less than .01); an… Show more

“…5B). Treatment of both Bov-LISN and Bov-LNL6 cultures with TGF-P resulted in 50% decreases in MyHC expression, in agreement with previous observations by Greene and Allen (1991) that TGF-P inhibits bovine myoblast differentiation. As in the dose-response curves generated without TGF-p, the IGF-I dose-response curve of Bov-LISN MyHC expression was more pronounced for both ascending and descending limbs compared to Bov-LNL6 cultures.…”

“…TGF-␤ represses skeletal muscle specific gene expression and has also been reported to modulate proliferation in satellite cells (3,12,19,24,47,59). Taken together, these studies indicate that TGF-␤ reprograms gene expression in muscle cells resulting in an alteration of proliferative control and a potent inhibition of the program of gene expression underlying myogenic differentiation.…”

“…The ability of TGF-b1 to inhibit myoblast differentiation and to depress proliferation has been documented (Massague et al, 1986;Allen & Boxhorn, 1987;Greene & Allen, 1991). More recently, human MPCs were cultured in the presence of 25 ng ml À1 human recombinant TGF-b1 (Carlson et al, 2009b).…”
